| 【目的】筛选高效纤维素降解菌,并对其产酶条件进行优化,为玉米秸秆等农业废弃物的工业化处理 和利用提供参考。【方法】使用纤维素-刚果红透明圈法,从玉米种植基地腐殖层土壤中分离具有高纤维素酶活性的 降解菌,通过形态学观察并结合分子生物学方法,对筛选出的菌株进行鉴定,然后对该菌株的营养条件和培养条件进 行优化。【结果】从土壤中分离获得一株纤维素降解菌ZZ04,经鉴定为贝莱斯芽孢杆菌(Bacillus velezensis),该菌株培养时的最适碳源为羧甲基纤维素钠(CMC-Na),氮源为蛋白胨与酵母粉的等量混合物,碳氮比为2∶4,接种量为3%,培养基初始pH值为7,培养温度为35 ℃,培养时间为48 h,在此条件下菌株ZZ04产羧甲基纤维素酶(CMC)活性和滤纸酶(FPA)活性分别高达222.14和171.21 U/mL。【结论】筛选的贝莱斯芽孢杆菌ZZ04在最佳培养条件下的纤维素 酶活性较高,具有开发微生物降解菌剂的潜力。 |

| 【Objective】This study aims to screen for efficient cellulose-degrading strains,and optimizes their enzyme production condition,thereby providing a reference for the industrial treatment and utilization of such agricultural waste as corn straw.【Method】Using the cellulose-Congo red clear zone method,strains with high cellulase activity were isolated from the humus layer soil of a corn planting base. The selected strains were identified by morphological observation combined with molecular biological analysis,followed by optimization of their nutritional and cultural conditions.【Result】A cellulose-degrading strain ZZ04 was isolated from soil and identified as Bacillus velezensis. The optimal culture conditions for this strain were as follows:CMC-Na as the carbon source,an equal mixture of peptone and yeast extract as the nitrogen source,a carbon-to-nitrogen ratio of 2∶4,an inoculation size of 3%,an initial medium pH of 7,a culture temperature of 35 ℃,and an incubation time of 48 h. Under these conditions,the carboxymethyl cellulase(CMC) activity and filter paper activity(FPA) of the strain reached 222.14 U/mL and 171.21 U/mL,respectively.【Conclusion】The screened strain B.velezensis ZZ04 exhibits high cellulase activity under optimal culture conditions,indicating its potential for development as a microbial degradation agent. |
