| ‘翠碧一号’不同部位鲜烟叶成熟过程中 代谢物差异分析 |

| 【目的】明确‘翠碧一号’(CB-1)成熟过程中上部叶和中部叶物质转化规律及代谢物差异,为‘翠碧一 号’上部叶和中部叶适宜采收成熟度的判断提供理论依据。【方法】设置尚熟(M1)、成熟(M2)和过熟(M3)3 个成熟梯度,利用液相色谱-串联质谱(LC-MS/MS),结合主成分分析(PCA)和正交偏最小二乘法-判别分析(OPLS-DA)对上部和中部烟叶代谢物进行分析。【 结果】不同成熟度鲜烟叶代谢物差异显著,上部叶和中部叶分别筛选出149和271种差异代谢物;上部叶的关键代谢通路为各种生物碱的生物合成,苯丙氨酸、酪氨酸和色氨酸的生物合成,色氨酸代谢及苯丙氨酸代谢;中部叶的关键代谢通路为 α-亚麻酸代谢、半胱氨酸和蛋氨酸代谢及抗坏血酸和醛糖酸代谢。3-吲哚丙烯酸、8-羟基-7-甲氧基香豆素、氧化苦参碱是‘翠碧一号’上部叶成熟过程中的关键代谢物,随烟叶采收成熟度 的提高,3-吲哚丙烯酸和氧化苦参碱相对含量呈逐渐上升趋势,8-羟基-7-甲氧基香豆素相对含量呈先上升后下降的 趋势;α-戊基肉桂醛、壬二酸、2-十二烯二酸是‘翠碧一号’中部叶的关键代谢物,三者相对含量随烟叶采收成熟度的提 高呈先上升后下降的趋势。【结论】3-吲哚丙烯酸、8-羟基-7-甲氧基香豆素、氧化苦参碱、α-戊基肉桂醛、壬二酸、2-十 二烯二酸可作为‘翠碧一号’烟叶成熟的潜在标志物。 |

| 【Objective】The study aims to clarify the patterns of material transformation and metabolite differences in the upper and middle leaves of CB-1 during maturation,providing a theoretical basis for determining the suitable harvesting maturity of upper and middle leaves in CB-1.【Method】Three maturation gradients were set:immature(M1),mature(M2),and over-mature(M3).Metabolites in upper and middle tobacco leaves were analyzed using liquid chromatography-tandem mass spectrometry(LC-MS/MS) combined with principal component analysis(PCA) and orthogonal partial least squares discriminant analysis(OPLS-DA).【Result】Significant differences in metabolites were observed in fresh tobacco leaves of different maturities. A total of 149 and 271 differential metabolites were screened from the upper and middle leaves,respectively. The key metabolic pathways in the upper leaves involved the biosynthesis of various alkaloids,biosynthesis of phenylalanine,tyrosine,and tryptophan,tryptophan metabolism,and phenylalanine metabolism. The key metabolic pathways in the middle leaves included α-linolenic acid metabolism,cysteine and methionine metabolism,and ascorbate and aldarate metabolism. 3-indolepropionic acid,8-hydroxy-7-methoxycoumarin,and oxymatrine were key metabo‐lites in the maturation process of the upper leaves of CB-1. With increasing maturity of the tobacco leaves,the relative contents of 3-indolepropionic acid and oxymatrine showed a gradual upward trend,while the relative content of 8-hydroxy-7-methoxycoumarin first increased and then decreased. α -pentylcinnamaldehyde,azelaic acid,and traumatic acid were key metabolites in the middle leaves,and their relative contents first increased and then decreased with increasing maturity of the tobacco leaves.【Conclusion】3-indolepropionic acid,8-hydroxy-7-methoxycoumarin,oxymatrine,α
-pentylcinnamaldehyde,azelaic acid,and traumatic acid may serve as poten‐tial markers for the maturation of CB-1 tobacco leaves. |
