| 【目的】探究盐胁迫对黄瓜(Cucumis sativus L.)和白籽南瓜(Cucurbita maxima×Cucurbita moschata)根系边缘细胞(root border cells,RBCs)的影响,评价黄瓜和白籽南瓜耐盐的细胞学机制。【方法】以‘津研四号’黄瓜和‘金甲砧’白籽南瓜种子为试验材料,采用浸种法和悬空培养法,研究0(对照),50,100,150和200 mmol/L NaCl溶液对其种子发芽率、根长、根鲜质量、根系活力以及根系边缘细胞数目、活率、黏胶层厚度和凋亡率的影响。【结果】随着NaCl浓度的增加,黄瓜和白籽南瓜种子的发芽率呈先升高后降低趋势,其中黄瓜种子的发芽率较白籽南瓜种子降低更明显;黄瓜和白籽南瓜的根长、根鲜质量和根系活力均呈下降趋势,且黄瓜的降低幅度较白籽南瓜更大。与对照相比,NaCl处理降低了黄瓜和白籽南瓜根系边缘细胞的数目和活率,且黄瓜降低幅度较白籽南瓜更明显;但NaCl处理却增加了黄瓜和白籽南瓜根系边缘细胞的黏胶层厚度和凋亡率。在NaCl浓度相同时,白籽南瓜根系边缘细胞的黏胶层厚度大于黄瓜,根系边缘细胞的凋亡率则小于黄瓜。【结论】根系边缘细胞(RBCs)能提高植物抵御NaCl胁迫的能力;白籽南瓜因具有较多的根系边缘细胞数目、较高的根系边缘细胞活率、较厚的黏胶层和较低的根系边缘细胞凋亡率,致使其抵御NaCl胁迫的能力强于黄瓜。 |

| 【Objective】The study aimed to explore the effects of salt stress on root border cells (RBCs) of cucumber (Cucumis sativus L.) and pumpkin (Cucurbita maxima×Cucurbita moschata),and to evaluate the cytological mechanisms of salt tolerance in the two melon species.【Method】The seeds of ‘Jinyan No.4’ cucumber and ‘Jinjia Anvil’ pumpkin were used as test materials to study the effects of 0 (control,CK),50,100,150 and 200 mmol/L NaCl on seed germination rate,root length,root fresh mass,root vitality,RBCs number,viability,mucilage layer thickness,and apoptosis rate using soaking method and suspension culture method.【Result】With the increase of NaCl concentration,the germination rate of the cucumber and pumpkin seeds showed a trend of increasing and then decreasing,with cucumber decreasing more significantly than pumpkin.The root length,root fresh mass and root viability of both cucumber and pumpkin showed a decreasing trend,and the decrease of cucumber was greater than that of pumpkin.Compared with the control group,NaCl treatment decreased the number and viability of RBCs in cucumber and pumpkin,and the decrease in cucumber was more obvious than that of pumpkin.In addition,NaCl treatment increased the thickness of the mucilage layer and apoptosis rate of RBCs in the two melons,and the thickness of the mucilage layer of RBCs in pumpkin was greater than that of cucumber at the same concentration of NaCl,while the apoptosis rate of RBCs was less than that of cucumber.【Conclusion】RBCs increased the ability of plants to resist NaCl stress.Pumpkin was stronger than cucumber in resisting NaCl stress due to a larger number of RBCs,higher RBCs viability,a thicker mucilage layer,and a lower apoptosis rate of RBCs. |
