甘肃冬小麦品种(系)抗条锈基因Yr18的鉴定及其转育

摘要:
【目的】了解抗锈基因Yr18在甘肃省冬小麦新品种(系)中的分布,验证标记的有效性,选育慢锈小麦新品种。【方法】以含Yr18基因的CP90-02-4-1为抗源,以洮157为农艺亲本组配杂交组合,培育小麦新品系,并利用分子标记csLV34和形态标记Ltn1对42份冬小麦新品种(系)、21份抗源亲本及洮157衍生后代进行Yr18基因鉴定。【结果】(1)csLV34有2种多态性,凡含有Yr18的材料均能扩增出150 bp的片段,不含Yr18的材料能扩增出229 bp的片段。但Ltn1对应的叶干尖不具有唯一性。(2)检测材料Yr18的分布频率低,主要原因是亲本携带目标基因的频率低。(3)以含慢条锈基因Yr18的材料为抗源,改良高产优质抗性丧失栽培种的可操作性强,是培育丰产、优质抗条锈小麦新品种的有效途径。【结论】标记csLV34可用于慢条锈基因Yr18的分子标记辅助选择,但叶干尖不能准确鉴别小麦品种(系)携带Ltn1;冬小麦新品种(系)Yr18的分布频率低,但Yr18的转育效率高。

Identification and transfer of Yr18 for resistance to stripe rust in Gansu winter wheat varieties (lines)

Abstract:
【Objective】The present study was to understand the distribution of Yr18 for resistance to stripe rust in Gansu winter wheat varieties lines,verify the availability of markers used,and breed new wheat lines with slow rusting gene.【Method】Progenies were produced with the resistant parent CP90-02-4-1 carrying Yr18 and agronomic parent Tao 157,and at the same time,the slow rusting gene Yr18 was tested in 42 Gansu winter wheat varieties (lines),21 resistant parents and descendants from Tao 157 to investigate the occurrence frequency and transferring efficiency of Yr18 with STS marker csLV34 and Ltn1,which could speed up the resistant breeding procedure.【Result】(1)csLV34 produced two polymorphisms,a 150 bp fragment amplified indicated the presence of Yr18,whereas a 229 bp fragment amplipied in the other genotypes indicated the absence of Yr18.The varieties with Ltn1 had leaf tip necrosis,but the varieties with leaf tip necrosis didn’t definitely contain Ltn1.(2) The frequency of materials with Yr18 was low,mainly due to few parents with the targeted gene.(3)It was effective for resistant variety with Yr18 being transferred to the susceptible but locally adapted varieties such as Tao 157,and this breeding way was a shortcut for breeding excellent wheat varieties with high yield,good quality and high resistance to stripe rust.【Conclusion】The co-dominant STS marker csLV34 was closely linked to Yr18,could be used as a tool for marker assisted selection of Yr18 in wheat variety improvement for resistance to stripe rust,but leaf tip necrosis could not accurately describe the presence or absence of Ltn1.The frequency of Yr18 in the materials tested was low and tranferring efficiency for Yr18 was relatively high.
