金丝桃苷对SLT-Ⅱ e诱导大鼠肠黏膜微血管内皮细胞分泌PGI2、TXA2及PAF的影响

摘要:
通过检测金丝桃苷对SLT-Ⅱe诱导的大鼠肠黏膜微血管内皮细胞分泌前列环素(PGI2)、血栓素(TXA2)及血小板活化因子(PAF)的影响,探索中药复方有效成分金丝桃苷对仔猪水肿病的疗效机制.将培养的肠黏膜微血管内皮细胞分为阴性对照组、阳性对照组、不同质量浓度金丝桃苷处理组等6个组,采用ELISA法测定了培养3,6,9和12 h时细胞上清液中PGI2、TXA2及PAF质量浓度的变化.结果表明,不同质量浓度金丝桃苷处理组对SLT-Ⅱe诱导肠黏膜微血管内皮细胞过量分泌PGI2、PAF无明显的抑制作用;1和5 μg/mL质量浓度金丝桃苷处理组对SLT-Ⅱe诱导肠黏膜微血管内皮细胞TXA2的分泌具有明显的下调作用.由此可见,金丝桃苷通过抑制SLT-Ⅱe诱导肠黏膜微血管内皮细胞TXA2的过量分泌,阻止血小板聚集,避免微血栓形成,以达到治疗疾病的目的;金丝桃苷保护大鼠肠黏膜微血管内皮细胞损伤的模型剂量为1和5μg/mL.

Effects of active various concentrations of hyperoside on PGI2,TXA2 and PAF production induced by SLT-Ⅱe in rat intestinal microvascular endothelial cells
Abstract:
The study observed the effects of various concentrations of hyperoside on PGI2,TXA2 and PAF production induced by SLT-Ⅱe in rat intestinal microvascular endothelial cells so as to probe curative effect mechanism of edema disease of swine. The RIMECs are divided into negative control group(0 μg/mL SLT-Ⅱe),masculine control group(10 μg/mL SLT-Ⅱe),test 3 group(10 μg/mL SLT-Ⅱe+1μg/mL hyperoside),test 4 group (10 μg/mL SLT-Ⅱe+5 μg/mL hyperoside),test 5 group(10 μg/mL SLT-Ⅱe +10 μg/mL hyperoside) and test 6 group(10 μg/mL hyperoside).In each group various concentrations of hyperoside against PGI2TXA2 and PAF production induced by SLT-Ⅱe in RIMECs were studied by ELISA. The results showed that hyperoside had inhibited the secretion of SLT-Ⅱe inducing PGI2,PAF production in RIMECs,but showed unconspicuous declining;while 1,5 μg/mL hyperoside had inhibited the secretion of SLT-Ⅱe inducing TXA2 production in RIMECs and showed marked declining. Hyperoside had inhibited the secretion of SLT-Ⅱe inducing TXA2 production there after to counterwork blood platelet conglomeration,avoid tiny thrombus form and heal disease;then the model concentration of hyperoside was 1,5 μg/mL to protect the RIMECs.
