| 采用杜仲优树的幼叶、嫩茎及种子无菌苗的下胚轴、子叶、真叶、根为外植体,以B5和MS为基本培养基,添加1.0 mg/L NAA和0.3 mg/L BA,进行了不同外植体种类、外植体大小、试验接种方式、外植体采样时间、光照条件、培养基pH值共6种因素对杜仲愈伤组织诱导的影响试验。结果表明,在杜仲愈伤组织诱导中,田间材料幼叶取样时间以3月中旬为好,幼茎的取材时间以4月中旬以前最佳;无菌苗以下胚轴、子叶、真叶诱导效果较好;诱导愈伤组织的培养基pH值以6.3较适宜;培养室的光照条件以12 h光照、12 h暗培养较有利于愈伤组织诱导;茎段的大小和接种方式以0.7 cm长度横放方式诱导效果较好。 |

| Abstract: |
| In callus induction of Eucommia ulmoides Oliv.,we used shoot,leaf of superior trees and hypocotyl,cotyledon,leaf,root of the sterile seedling as explants.Factors affecting callus induction of the explants of type,length,ways of implanting,sampling time,light and pH were studied on MS and B5 medium supplemented with 1.0 mg/L NAA and 0.3 mg/L BA.The results showed that,middle of March proved to be a favorite time for immature leaves as explants.Before middle April was the time suitable for shorts.Hypocotyl,cotyledon and leaf of sterile seedling were best.The best pH for the callus induction was 6.3.12 hour light and 12 hour dark was benefit for the callus induction.0.7 cm shoot explants implanted horizontally in the medium was best. |
