通过植株原位真空渗入遗传转化获得转基因芥菜

摘要:
以芥菜(Brassica juncea Coss)雪里蕻和圆叶芥为试材,在携带潮霉素磷酸转移酶基因和CaMV Bari-1基因Ⅵ的根癌农杆菌菌株GV3101的介导下,采用植株原位真空渗入遗传转化技术,将外源基因潮霉素磷酸转移酶基因和CaMV Bari-1基因Ⅵ导入芥菜。采收T1种子,经潮霉素抗性筛选和PCR分子检测,证实获得了转基因植株。

Transformation of mustards (Brassica juncea Coss.) by the plant in situ vacuum infitration
Abstract:
Mustards (Brassica juncea Coss.) ecotype Xuelihong and Yuanyejie were transformed with in planta vacuum infiltration by an Agrobacterium tumefaciens strain GV3101 carrying hygromycin resistance gene and CaMV Bari-1 gene Ⅵ.The T1 seeds of mustards were screened through medium with 25 mg/L hygromycin and some resistant seedlings were obtained.PCR molecular analyses showed that target genes were transferred to the plant cells.
