植物afp表达载体的构建及其对农杆菌的直接转化

摘要:
详细报道了胡萝卜抗冻蛋白基因(afp)植物表达载体的构建过程及其直接法转化农杆菌的方法。以限制性内切酶从克隆载体上切取afp基因,将其连接在相应酶切的pBI121工程质粒上,构建成afp植物表达载体。采用直接转化法将重组子导入根癌农杆菌,并用PCR方法进行了鉴定。

Construction of plant afp expression vector and its direct transformation to agrobacterium
Abstract:
Construction of a vector expressing carrot antifreeze protein gene(afp) in plants and its direct transformation to agrobacterium were thoroughly reported.Clone vector was digested with restriction endonucleases,and the acquired afp was ligated to engineered plasmid p BI121 which was digested with appropriate restriction endonucleases,and therefore a plant expression vector for carrot afp was constructed.The recombinant was transferred into agrobacterium tumefacien by direct DNA transfer and the transformant was identified with PCR.
