| 茯苓胞外三萜与甾醇分析及其抗氧化活性发掘 |

| 【目的】研究茯苓胞外三萜和胞外甾醇化合物的组成并分析其抗氧化活性,为茯苓菌株质量控制和抗氧化产品开发提供参考。【方法】以不同来源的FLA、FGD、FGZ、FHB、GTR1、GTR2、XJ28、F5.78等8个茯苓菌株为研究对象,利用广泛靶向代谢组学分析茯苓胞外三萜和胞外甾醇的化学成分,利用ABTS、DPPH、FRAP和OH模型分析各茯苓菌株发酵液的抗氧化能力,并利用聚类分析、主成分分析、正交偏最小二乘法判别分析和Pearson相关性分析,确定8个茯苓菌株胞外三萜和胞外甾醇的差异成分。【结果】从8个茯苓菌株中共定性检测出24种化合物,包括18种胞外三萜类化合物和6种胞外甾醇类化合物。其中GTR1菌株相对含量最高的三萜类成分是灵芝酸N,其他7个菌株相对含量最高的三萜类成分均为齐墩果酸甲酯,而菜油甾-4-烯-3β醇是8个菌株相对含量最高的甾醇类成分。GTR2菌株在ABTS、DPPH和FRAP模型中均显示出较强的抗氧化能力,而XJ28菌株在OH模型中显示出极强的抗氧化能力。正交偏最小二乘法判别分析发现,8个茯苓菌株的胞外三萜和胞外甾醇中存在10个差异组分,包括8个三萜类化合物和2个甾醇类化合物。Pearson相关性分析发现,ABTS、FRAP、DPPH模型分别与24,24,22种化合物峰面积相对含量呈负相关,而OH模型与12种化合物峰面积相对含量呈正相关。10个差异组分中,3-表熊果酸、去氢茯苓酸的相对含量与DPPH模型呈显著负相关(P<0.05),茯苓新酸B的相对含量与ABTS模型呈显著负相关(P<0.05),灵芝酸S、3β-羟基麦角甾-7,24(24(1))-二烯-4α-羧酸酯、4α-甲基-5α-麦角甾-8,14,24(28)-三烯-3β醇的相对含量与OH模型呈极显著正相关(P<0.01)。【结论】不同茯苓菌株胞外三萜和胞外甾醇类化合物组成存在差异,3-表熊果酸、去氢茯苓酸、茯苓新酸B、灵芝酸S、3β-羟基麦角甾-7,24(24(1))-二烯-4α-羧酸酯、4α-甲基-5α-麦角甾-8,14,24(28)-三烯-3β醇是影响其抗氧化能力的关键成分。 |

| 【Objective】The composition of extracellular triterpenoids,extracellular sterols,and the antioxidant activities in Wolfiporia hoelen were studied in order to provide reference for quality control and antioxidant product development of W.hoelen strains.【Method】8 W.hoelen strains from different sources were studied,namely FLA,FGD,FGZ,FHB,GTR1,GTR2,XJ28,and F5.78.Widely targeted metabolomics was used to analyze the chemical components of extracellular triterpenoids and sterols.ABTS model,DPPH model,FRAP model,and OH model were used to determine the antioxidant capacity of fermentation broth.Cluster analysis,principal component analysis,orthogonal partial least squares-discriminant analysis,and Pearson correlation analysis were used to determine the differential components of extracellular triterpenoids and sterols in 8 W.hoelen strains.【Result】24 compounds were qualitatively detected in 8 W.hoelen strains,including 18 extracellular triterpenoids and 6 extracellular sterols.The triterpenoid component with the highest relative content in GTR1 strain was ganoderic acid N,while triterpenoid component with the highest relative content in the other seven strains was methyl oleanonate.For all 8 strains,the sterol component with the highest relative content was campest-4-en-3β-ol.The GTR2 strain of W.hoelen showed strong antioxidant capacity in the ABTS model,DPPH model,and FRAP model,while the XJ28 strain showed extremely strong antioxidant capacity in the OH model.According to orthogonal partial least squares-discriminant analysis,10 differential components were identified in 8 W.hoelen strains,including 8 triterpenoids and 2 sterols.Pearson correlation analysis showed that ABTS,FRAP and DPPH models were negatively correlated with the relative peak area content of 24,24 and 22 compounds,respectively,while OH model was positively correlated with the relative peak area content of 12 compounds.Among the 10 differential components,the relative contents of 3-epiursolic acid and dehydropachymic acid were only significantly negatively correlated with DPPH model (P<0.05).The relative content of poricoic acid B was only significantly negatively correlated with ABTS model (P<0.05).The relative contents of ganoderic acid S,3β-hydroxyergosta-7,24(24(1))-dien-4α-carboxylate, 4α-methyl-5α-ergosta-8,14,24(28)-trien-3β-ol were only significantly positively correlated with OH model (P<0.01).【Conclusion】There were differences in the composition of extracellular triterpenoids and sterols among different W.hoelen strains.3-epiursolic acid,dehydropachymic acid,poricoic acid B,ganoderic acid S,3β-hydroxyergosta-7,24(24(1))-dien-4α-carboxylate and 4α-methyl-5α-ergosta-8,14,24(28)-trien-3β-ol were key components affecting the antioxidant capacity. |
