| 【目的】解析牛肌肉发育过程中超级增强子、潜在靶基因及转录因子的调控机制。【方法】利用牛原代成肌细胞H3K27ac和H3K4me1 2种组蛋白修饰的ChIC-seq数据,分析和鉴定牛原代成肌细胞基因组超级增强子。利用ROSE软件注释超级增强子的靶基因,并对其进行GO和KEGG分析。对超级增强子区域进行基序富集分析。【结果】利用H3K27ac标记鉴定得到295个超级增强子,利用H3K4me1标记鉴定出362个超级增强子,其中二者共有超级增强子为143个。GO和KEGG分析表明,共有超级增强子调控的潜在靶基因主要富集在黏着斑、PI3K-Akt和病毒感染等信号通路,这些通路在调控成肌细胞增殖、分化和肌纤维形成中发挥着关键作用。对共有超级增强子区域进行基序富集分析,筛选并确定了MYB70和HLF等肌肉发育关键转录因子。【结论】利用2种组蛋白修饰筛选并鉴定出牛原代成肌细胞基因组内的超级增强子,为解析牛肌肉发育过程中超级增强子的结构、功能及调控机制提供了理论基础。 |

| 【Objective】This study provided a theoretical basis for understanding the regulatory mechanisms of super enhancers,putative target genes and transcription factors during bovine muscle cells development.【Method】ChIC-seq data of H3K27ac and H3K4me1 were successfully applied to identify SEs in primary bovine muscle cells.ROSE software was used to annotate the target genes of SEs and GO and KEGG analyses on these genes were subsequently conducted.Furthermore,motif enrichment analysis was performed on the regions of super-enhancers.【Result】295 SEs were identified by H3K27ac mark,while 362 were identified by H3K4me1 mark,among which 143 SEs were marked by both histone modifications.GO and KEGG analyses showed that the putative target genes,regulated by shared SEs,mainly involved in focal adhesion,PI3K-Akt signaling and viral infection pathways,which played crucial roles in regulating muscle cell proliferation,differentiation,and myofiber formation processes.Through further enrichment analysis of transcription factor binding motifs in shared SEs regions,MYB70 and HLF were screened out and identified as key transcription factors during muscle development.【Conclusion】This study identified the distribution of super enhancers with two histone modification approaches,laying a theoretical foundation for deciphering the structure,function,and regulatory mechanisms of super enhancers during bovine muscle development. |
