航天诱变秀珍菇菌株的特性研究

摘要:
【目的】研究航天诱变所得秀珍菇优势变异菌株的多态性及胞内活性成分含量,为秀珍菇育种及其附加价值开发提供参考。【方法】2020年5月对秀珍菇进行航天诱变,以菌丝生长速度为指标筛选优势变异株,以出发菌株为对照,对优势变异株进行ITS鉴定和拮抗试验;利用ISSR、RAPD、SRAP分子标记研究优势变异株的多态性和遗传变化,测定其胞内活性成分含量;运用POPGENE 32和NTSYS PC V2.10软件对各菌株进行遗传变异和聚类分析。【结果】获得52株秀珍菇航天诱变再生菌株,且有25株菌丝生长速度变化显著;其中有6株表现优异,且与出发菌株存在明显的拮抗现象。ISSR、RAPD、SRAP的18条引物在6株优势变异株中共扩增出62条带,多态性条带占比72.58%。遗传变异分析显示,优势变异株群的Nei’s基因多样性指数为0.269 8,Shannon’s多态性信息指数为0.399 1;基于ISSR+RAPD+SRAP的数据显示,出发菌株与6优势变异株间的遗传相似性系数为0.629 0~0.871 0,6株优势变异株间的遗传相似性系数为0.467 7~0.774 2。对出发菌株和6株优势变异株进行聚类分析,在遗传距离0.62处可分为2类,说明秀珍菇优势变异菌株在遗传水平上发生了变异。最终筛选到3株总酚、黄酮含量和2株黄酮、多糖含量同时提高的菌株,其活性成分含量较出发菌株显著提高了20.30%~122.34%。【结论】航天诱变后,秀珍菇菌株发生较大变异,并筛选出5株特性优良的菌株,其活性成分含量较高。

Characterization of Pleurotus geesteranus strains induced by space flight

Abstract:
【Objective】This study investigated the polymorphisms and intracellular active component contents of dominant variant strains of Pleurotus geesteranus induced by space mutation to provide references for breeding and increasing added values.【Method】In May 2020,spaceflight mutagenesis was carried out on Pleurotus geesteranus,and dominant variant strains were screened by the hypha growth rate.ITS analysis and antagonistic experiment were performed for dominant variant strains using the starting strain as control.ISSR,RAPD and SRAP were used to study the polymorphisms and genetic variations in dominant variant strains,and the intracellular active ingredients were determined.The POPGENE 32 and NTSYS-PC V2.10 were used to analyze genetic variations.【Result】Among the 52 regeneration strains of Pleurotus geesteranus induced by space mutation,25 strains showed significantly different growth rates and 6 strains showed excellent growth rates with antagonistic phenomenon against the starting strain.A total of 62 bands were amplified by 18 primers for ISSR,RAPD and SRAP in the 6 dominant mutant strains,of which the proportion of polymorphism was 72.58%. Genetic variation analysis showed that Nei’s gene diversity index was 0.269 8 and Shannon’s polymorphism information index was 0.399 1.The ISSR+RAPD+SRAP based data showed that the genetic similarity coefficients between the starting strain and the dominant mutant strains were 0.629 0 to 0.871 0,and the genetic similarity coefficients between the dominant mutant strains were 0.467 7 to 0.774 2.Cluster analysis of the starting strains and dominant variants showed that they could be classified into two classes at a genetic distance of 0.62,indicating variation in the genetic level.Three strains with simultaneously increased total phenolics and flavone contents and two strains with simultaneously increased flavone and polysaccharide contents were screened,and the contents of active components were increased by 20.30%-122.34%.【Conclusion】After space mutation,Pleurotus geesteranus showed considerable variation and five strains with high active ingredients were selected.
