柿种质资源果实糖组分及含量多样性分析

摘要:
【目的】探索不同柿种质资源成熟果实糖组分及构成比例,为柿资源的合理保存、有效利用提供理论依据。【方法】以41份柿种质资源成熟果实及1份CO2脱涩处理的硬实期平核无果实为试材,采用高效液相色谱法测定果肉中果糖、山梨醇、葡萄糖和蔗糖含量,并采用离散特征分析、相关性分析和聚类分析等方法研究柿果实糖组分含量的多样性。【结果】41份柿种质资源均包含4种糖组分,其中山梨醇和蔗糖含量较低,果糖和葡萄糖含量较高,且各糖组分占总糖比例差异较大。相关性分析表明,总糖含量与4种糖组分含量均呈显著或极显著正相关,山梨醇与果糖、葡萄糖和蔗糖含量呈极显著正相关,甜度值与总糖、果糖和葡萄糖含量呈极显著正相关。聚类分析结果显示,将41份柿种质在遗传距离为15处划分为Ⅰ、Ⅱ、Ⅲ 3个类群,在遗传距离为5处将第Ⅰ类群划分为Ⅰ-a和Ⅰ-b亚类,其中Ⅰ-b亚类为果糖积累型,其余类群柿种质均为己糖积累型。基于糖组分及含量的多样性分析,筛选出了总糖含量较高的种质有休宁扁塌柿、暑黄柿和甜宝盖,甜度值较高种质有休宁扁塌柿和蛭干八月黄,并发现CO2脱涩处理的平核无果实中果糖、葡萄糖、总糖和甜度值均明显高于自然软熟的果实。【结论】柿种质资源糖含量及构成比例多样性丰富,筛选出的特异资源为以风味多样化为目标的新品种杂交育种提供了亲本。

Components and contents diversity of soluble sugars in mature fruit of persimmon germplasm

Abstract:
【Objective】This study explored components and composition of soluble sugars in different persimmon germplasm resources to provide basis for rational conservation and effective utilization of persimmon resources.【Method】Fructose,sorbitol,glucose and sucrose contents in 41 mature fruits of persimmon germplasm resources and one hard-ripened persimmon fruit treated with CO2 desiccation were determined using high performance liquid chromatography.The diversity of sugar contents of persimmon fruit was investigated using dispersion analysis,correlation analysis and cluster analysis.【Result】All 41 persimmon germplasm resources contained four sugar components.Sorbitol and sucrose contents were low,fructose and glucose contents were high,and their ratios to total sugar differed greatly.Correlation analysis showed that total sugar was significantly and positively correlated with all four sugar components,sorbitol was significantly and positively correlated with fructose,glucose and sucrose,and sweetness values were significantly correlated with total sugars,fructose and glucose.The clustering analysis divided the 41 persimmon germplasm into three groups at a genetic distance of 15,and the first group was divided into subgroups Ⅰ-a and Ⅰ-b at a genetic distance of 5.Subgroup Ⅰ-b was fructose accumulating,while the rest was hexose accumulating.Based on the diversity analysis of sugar components and contents,Xiuning Biantashi,Shu Huangshi and Tian Baogai with high total sugar content as well as Xiuning Biantashi and Zhigan Bayuehuang with high sweetness value were selected.The fructose,glucose,total sugars and sweetness values of the CO2-treated fruit were significantly higher than those of naturally soft-ripened fruits.【Conclusion】The diversity of persimmon sugar content and composition was rich and the selected specific resources could provide parents for crossbreeding new species with flavor diversification.
