酸枣经济性状关联分析及优异等位基因挖掘

摘要:
【目的】探寻与酸枣主要经济性状相关联的SSR位点,为酸枣优良性状基因挖掘、优良亲本选育及资源开发利用提供理论依据。【方法】以辽西地区的72份酸枣种质为研究对象,在成熟期调查其单果质量、单核质量、单仁质量、果实横径、果实纵径、果核横径、果核纵径、果仁横径、果仁纵径、果仁侧径、果形指数、核形指数、仁形指数、出核率、出仁率、双仁率及可食率,利用SPSS 22.0软件对上述17个经济性状进行统计分析;选用32个SSR标记,利用Structure 2.3.4软件分析72份酸枣种质的群体结构;利用Tassle 2.0软件进行32个SSR标记间的连锁不平衡分析,采用一般线性模型(GLM)和混合线性模型(MLM)相结合的方法进行各经济性状与SSR标记间的关联分析,并对关联位点等位变异的表型效应进行分析,从而获得典型载体材料。【结果】72份酸枣种质17个经济性状的变异系数为7.84%~85.75%,表明供试种质具有较为丰富的表型多样性;通过群体遗传结构分析将72份酸枣种质分为4个类群,说明群体遗传背景较为单一。32对SSR分子标记间的连锁不平衡水平较低,成对存在的不平衡组合有166个,占组合总数的33.47%。在关联分析中,通过GLM模型检测出21个SSR位点与17个主要经济性状显著相关联(P<0.05),变异解释率为15.35%~69.14%;通过MLM模型检测出17个SSR位点与16个经济性状显著相关联,变异解释率为0.82%~75.37%。对2种模型中共同检测到的16个SSR关联位点各等位片段的表型效应值进行分析,共挖掘出31个具有最大增效与最大减效的优异等位变异和15个典型载体材料。【结论】基于SSR的关联分析结果,筛选出与酸枣16个经济性状相关联的16个SSR关联位点及15个聚合优异等位变异的典型载体材料。

Association analysis of economic traits and mining of excellent alleles of Ziziphus jujuba var. spinosa

Abstract:
【Objective】This study explored SSR loci associated with main economic characters of Ziziphus jujuba var.spinosa to provide basis for gene mining, excellent parent breeding and resource exploitation of Z.jujuba var.spinosa.【Method】The single fruit mass,fruit horizontal diameter,fruit vertical diameter,fruit-form index,single nutlet mass, nutlet horizontal diameter,nutlet vertical diameter,nucleation index,single kernel mass,kernel horizontal diameter,kernel vertical diameter,kernel-form index,kernel side diameter,dual kernel rate,edible rate,nutlet rate and kernel rate of 72 Z.jujuba var.spinosa germplasms in western Liaoning were investigated.SPSS 22.0 software was used for statistical analysis of economic traits,32 SSR markers were selected, and Structure 2.3.4 software was used to analyze the population structure of germplasms.Tassle 2.0 software was used to analyze the linkage disequilibrium among 32 SSR markers.General linear model (GLM) and mixed linear model (MLM) were used to analyze the association between economic traits and SSR markers,and the phenotypic effects of alleles at association sites were analyzed to obtain typical vector materials.【Result】The variation coefficients of the 17 economic traits of 72 Z.jujuba var.spinosa germplasms ranged from 7.84% to 85.75%,indicating that the tested germplasms had rich phenotypic diversity.The 72 Z.jujuba var.spinosa germplasms were divided into 4 groups by population genetic structure analysis,indicating unique population genetic background.The level of linkage disequilibrium among 32 SSR markers was low.There were 166 pairs of unbalanced combinations,accounting for 33.47% of the total number of combinations.In the correlation analysis,21 SSR loci significantly associated with 17 major economic traits were detected by the GLM model (P<0.05),and the explained rate of variation was 15.35%-69.14%.The MLM model detected 17 SSR loci significantly associated with 16 economic traits and the explained rate of variation was 0.82%-75.37%.The phenotypic effect values of each allelic fragment of 16 SSR association sites detected in the two models were analyzed,and 31 excellent allelic variations with maximum efficiency increase and reduction and 15 typical carrier materials were excavated.【Conclusion】Based on SSR correlation analysis,16 SSR association site and 15 aggregated excellent alleles associated with 16 economic traits were selected.
