LPS对肉鸡不同组织lncRNA表达的影响

摘要:
【目的】探讨长链非编码RNA (lncRNA)在脂多糖(LPS)诱导的肉鸡系统性炎症反应中的表达情况及与促炎细胞因子白介素1β (IL-1β) 和白介素6 (IL-6)基因表达的相关性。【方法】选取21日龄Ross肉鸡10只,随机平均分为LPS组和对照组(CK),分别腹腔注射0.5 mg/kg LPS及相应体积生理盐水,于处理后2 h采集肌肉、肝脏及脾脏组织备用。以β-actin为内参基因,采用实时荧光定量PCR(RT-qPCR)检测3种组织样品中IL-1β、IL-6 mRNA的相对表达量。依据已报道的数据,选择肉鸡14个高表达lncRNA,采用RT-qPCR检测其相对表达量,并分析其与IL-1βIL-6基因mRNA表达量的相关性。【结果】LPS处理后2 h,肉鸡各组织中IL-1βIL-6基因mRNA表达量均极显著高于CK组。与CK组相比,LPS组肉鸡肌肉中lnc0035、lnc0181和TCO873的相对表达量显著或极显著升高,lnc0119和lnc0151的相对表达量显著或极显著降低;肝脏中lnc0035、lnc0181、lnc0202、TCO354、TCO873和pouBW1的相对表达量显著或极显著升高,lnc0119、lnc0151、TCO501和lncDHCR24的相对表达量显著或极显著降低;脾脏中lnc0181、lnc0202、TCO619和TCO873的相对表达量显著升高,lnc0035、lnc0119、lnc0128和lncDHCR24的相对表达量显著或极显著降低。lnc0035、lnc0119、lnc0181、TCO873 4种lncRNA在3种组织中的表达量与IL-1β、IL-6 mRNA表达量的相关性分析结果显示,肌肉中,除lnc0035与IL-6 mRNA表达量无显著相关性外,其他3个lncRNA均与IL-1βIL-6 mRNA的表达量呈显著或极显著相关;肝脏中,lnc0035、TCO873与IL-1βIL-6的mRNA表达量均呈显著正相关;脾脏中,TC0873表达与IL-1βIL-6的mRNA表达量呈极显著正相关,其余3个lncRNA均与IL-6 mRNA表达量呈显著或极显著相关。【结论】LPS能诱导肉鸡不同组织中多个lncRNA的表达产生差异,且lncRNA的表达与促炎细胞因子的IL-1βIL-6基因表达存在显著或极显著相关性,提示lncRNA可能参与了LPS诱导的全身性炎症反应。

Effect of LPS on lncRNAs expressions in different chicken tissues

Abstract:
【Objective】This study aimed to investigate the expression of long non-coding RNA (lncRNA) in lipopolysaccharide (LPS) induced systemic inflammatory response of broilers and its correlation with the expression of pro-inflammatory cytokine IL-1β and IL-6 genes.【Method】The 21-day-old Ross broilers were randomly divided into LPS group and control group (CK).Muscle,liver and spleen tissues were collected two hours after the intra-abdominal injection of 0.5 mg/kg LPS and corresponding volume of normal saline.Using β-actin as reference gene,the expression levels of IL-1β and IL-6 mRNA in three tissues were detected by real-time PCR.According to previous studies,14 high expressed lncRNAs in chicken were selected for detection by real-time PCR.The correlation between expressions of lncRNAs and IL-1β and IL-6 mRNA was also analyzed.【Results】Two hours after LPS treatment,the mRNA expressions of IL-1β and IL-6 in the three tissues were significantly higher than those in the control group.Compared with CK group,the expression levels of lnc0035,lnc0181 and TCO873 in muscle of broilers in LPS group were significantly or extremely significantly increased,while the expression levels of lnc0119 and lnc0151 were significantly or extremely significantly decreased.The expression levels of lnc0035,lnc0181,lnc0202,TCO354,TCO873 and pouBW1 in liver were significantly or extremely significantly increased,while the expression levels of lnc0119,lnc0151,TCO501 and lncDHCR24 were significantly or extremely significantly decreased.The expression levels of lnc0181,lnc0202,TCO619 and TCO873 in spleen were significantly increased,while the expression levels of lnc0035,lnc0119,lnc0128 and lncDHCR24 were significantly or extremely significantly decreased.The expressions of lnc0119,lnc0181 and TCO873 were significantly associated with IL-1β and IL-6 in muscle tissue,while the expression of lnc0035 only showed significant correlation with mRNA expression of IL-1β.Hepatic lnc0035 and TCO873 were significantly positively correlated with IL-1β and IL-6 mRNA expression.Only TCO873 expression in spleen was significantly positively correlated with IL-1β and IL-6 mRNA expression,while the rest were associated with IL-6 expression only.【Conclusion】LPS induced differential lncRNA expression in different tissues.The expressions of lncRNAs and pro inflammatory cytokines IL-1β and IL-6 had significant correlation,indicating that lncRNAs may participate in the systemic inflammatory response induced by LPS.
