| 【目的】对草原1号杂花苜蓿花蕾转录组微卫星SSR特征进行分析,为开发新的分子标记和辅助育种选择研究奠定基础。【方法】利用MISA软件对草原1号杂花苜蓿花蕾转录组数据进行简单重复序列(SSR)位点搜索,并对SSR分布及其序列特征进行分析。【结果】在286 409条非冗余单基因簇(Unigene)序列中共挖掘到 57 368个SSR位点,SSR出现频率为20.03%,平均分布距离5.84 kb。重复基元数量以单核苷酸(68.45%)为主,三核苷酸(16.46%)次之。在129种重复基元中,以A/T类型基元(68.15%)最高,其次为AG/CT基元(6.56%)。SSR基元重复次数主要集中在6~15次,核苷酸数量随着基元重复次数的增加而减少。SSR长度主要集中在12~40 bp(55.58%),具有较高多态性潜能的SSR(长度≥20 bp)位点数为9 166(15.98%)。【结论】草原1号杂花苜蓿SSR位点出现频率高、基元重复次数和类型丰富、多态性潜能高,可用于苜蓿遗传多样性与品种的分子鉴定及分子标记辅助选择。 |

| 【Objective】This study analyzed the SSR characteristics of buds transcriptome of Medicago varia Martin.‘Caoyuan No.1’ to provide basis for development of new molecular markers and assisted breeding selection.【Method】The MISA was used to explore all SSR loci in the transcriptome database of M. varia Martin.‘Caoyuan No.1’ and the SSR characteristics were analyzed.【Result】A total of 286 409 Unigenes were obtained from the buds transcriptome of M. varia Martin.‘Caoyuan No.1’,and 57 368 SSR loci were distributed with a frequency of 20.03% and a distribution density of 5.84 kb.Most repeat motifs were mono nucleotide motifs (68.45%) followed by tri nucleotide motifs (16.46%).The dominant repeat type was A/T (68.15%) among 129 repeat motifs and the second was AG/CT (6.56%).The motify repeats of SSR mainly concentrated in 6-15 times and decreased as the number of motif repetitions increases.The length of SSR was mainly 12-40 bp (55.58%) and the number of SSR (length ≥20 bp) with high polymorphic potential was 9 166 (15.98%).【Conclusion】The SSR locus in the buds transcriptome of M. varia Martin.‘Caoyuan No.1’ had high occurring frequency,abundant motif repetitions and types,and high polymorphism potential.These characteristics have great application potential in alfalfa genetic diversity,molecular identification of varieties and molecular marker assisted selection. |
