| 【目的】研究山茱萸叶多糖(PFC)的理化性质和单糖组成,分析其体外抗氧化活性及其对H2O2诱导的HepG2细胞氧化损伤的保护作用,为山茱萸资源的开发和利用提供理论依据。【方法】以干燥的山茱萸叶为原料,粉碎后经过水提醇沉、除蛋白、脱色等步骤后得到PFC;测定其总糖、糖醛酸、可溶性蛋白质和硫酸根含量;1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生化高效液相色谱法分析其单糖组成;测定PFC 对1,1-二苯基-2-三硝基苯肼自由基(DPPH·)、羟自由基(·OH)的清除能力及亚铁离子络合能力;检测不同质量浓度PFC溶液对H2O2诱导的HepG2细胞氧化损伤的保护作用,测定细胞内丙二醛(MDA)和活性氧(ROS)水平。【结果】PFC含总糖(612.76±1.72) mg/g,糖醛酸(228.03±0.54) mg/g,可溶性蛋白质(9.53±0.38) mg/g,硫酸根(25.21±0.19) mg/g。PFC由甘露糖、鼠李糖、半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖、岩藻糖构成,其物质的量比依次为1.6∶3.2∶6.2∶5.0∶13.0∶14.1∶1.0。PFC对DPPH·的IC50值为0.13 mg/mL,与对照维生素C(IC50为0.15 mg/mL)相当;对·OH的IC50值为0.56 mg/mL,低于对照维生素C(IC50为0.76 mg/mL)。PFC溶液可明显提高经H2O2诱导的HepG2细胞的相对存活率,降低其胞内MDA和ROS水平,对氧化损伤HepG2细胞具有保护作用,且呈剂量效应。【结论】山茱萸叶多糖具有良好的抗氧化活性,在保健食品和生物药品领域具有开发前景。 |

| 【Objective】This study investigated the physicochemical properties and monosaccharide composition of polysaccharide from Fructus corni (PFC) leaves and analyzed its antioxidant activities in vitro and protective effect on H2O2 induced oxidative damage of HepG2 cells to provide basis for the use of Fructus corni.【Method】Dried leaves of Fructus corni were crushed and extracted with water.After alcohol precipitation,protein removal and decolorization,the PFC was obtained.The contents of carbohydrate,uronic acid,soluble protein and sulfate radical were determined by colorimetry.The monosaccharide composition was measured as 1-phenyl-3-methyl-5-pyrazolone derivatives by high performance liquid chromatography.The scavenging activities against DPPH· radical and hydroxyl radical and the chelating ability to ferrous ion were measured.The protective effect of PFC solution with different mass concentrations on H2O2 induced oxidative damage of HepG2 cells was detected,and the cellular levels of malondiadehycle (MDA) and reactive oxygen specisis (ROS) were also determined.【Result】PFC contained carbohydrate (612.76±1.72) mg/g,uronic acid (228.03±0.54) mg/g,soluble protein (9.53±0.38) mg/g and sulfate radical (25.21±0.19) mg/g.PFC was composed of mannose,rhamnose,galacononic acid,glucose,galactose,arabinose and fucose in the molar ratio of 1.6∶3.2∶6.2∶5.0∶13.0∶14.1∶1.0.The 50% inhibitions (IC50) of PFC for DPPH· was 0.13 mg/mL,which was comparable to VC (0.15 mg/mL).The IC50 of PFC for ·OH was 0.56 mg/mL,lower than VC (0.76 mg/mL).PFC solution could concentration-dependently increase the cell relative survival rate and decrease intracellular MDA and ROS levels in H2O2 treated HepG2 cells.【Conclusion】The polysaccharide from Fructus corni leaves has good antioxidant capacity and can be a resource of health foods and biological drug for further development. |
