杂色云芝酸溶性多糖对H22荷瘤小鼠的抗肿瘤作用

摘要:
【目的】研究杂色云芝酸溶性多糖对H22荷瘤小鼠的抗肿瘤作用及可能的作用机制。【方法】首先测定杂色云芝酸溶性多糖内中性糖、葡萄糖醛酸及蛋白含量,然后以H22肝癌移植瘤小鼠为模型,设置空白对照组(等量生理盐水)、模型组(等量生理盐水)、环磷酰胺(CTX)组(30 mg/kg)和杂色云芝酸溶性多糖高、中、低剂量组(280,140,70 mg/kg),每天给药1次,连续给药10 d,比较各组抑瘤率、脏器指数、白细胞介素-6(IL-6)、γ-干扰素(INF-γ)、肿瘤坏死因子-α(TNF-α)、谷丙转氨酶(ALT)、尿素氮(BUN)和肌酐(CRE)等指标;最后在光镜下观察各处理组肿瘤组织H&E染色后的形态变化和细胞凋亡情况,考察杂色云芝酸溶性多糖的抗肿瘤作用及机制。【结果】杂色云芝酸溶性多糖内中性糖含量为656 mg/g,葡萄糖醛酸含量为38 mg/g,蛋白含量为18 mg/g。CTX组和杂色云芝酸溶性多糖高、中、低剂量组均能不同程度抑制肿瘤细胞增殖,抑瘤率分别为68.14%,55.56%,47.01%和37.52%;与CTX组相比,杂色云芝酸溶性多糖组改善了免疫器官指数(P<0.01);与模型组相比,杂色云芝酸溶性多糖组改善提高了血清细胞因子IFN-γ、TNF-α水平(P<0.01),降低了血清IL-6、ALT、BUN和CRE水平(P<0.01)。肿瘤组织H&E染色及凋亡检测结果表明,杂色云芝酸溶性多糖可促使肿瘤细胞凋亡。【结论】杂色云芝酸溶性多糖具有抗肿瘤作用,且可能是通过调节免疫能力实现这种作用的。

Antitumor effect of acid soluble polysaccharides from Trametes versicolor (L.) Lloyd in H22 tumor-bearing mice

Abstract:
【Objective】This paper aimed to study the antitumor effect and possible mechanism of acid soluble polysaccharides from Trametes versicolor(L.) Lloyd on H22 tumor-bearing mice.【Method】The contents of glucose,glucuronic acid and protein of the acid-soluble polysaccharides from Trametes versicolor(L.) Lloyd were determined and several groups including normal group (equal volume of saline),model group,(equal volume of saline),cyclophosphamide (CTX) group (30 mg/kg) as well as high,medium and low dose groups of acid-soluble Polysaccharides from Trametes versicolor (L.) Lloyd,acid-soluble polysaccharides were set with H22 hepatoma transplanted tumor mice as the model.The groups were treated once a day for 10 continuous days,and inhibitory rate,organ indexes of liver,thymus and spleen,and levels of IL-6,IFN-γ,TNF-α,ALT,BUN and CRE were measured and compared.The morphological changes after the H&E staining of tumor tissue and apoptosis were also observed under light microscope.【Result】The content of glucose in the acid-soluble polysaccharides from Trametes versicolor(L.) Lloyd was 656 mg/g,the content of glucuronic acid was 38 mg/g, and the content of protein was 18 mg/g.CTX group and the high, medium and low groups of acid soluble polysaccharides from Trametes versicolor (L.) Lloyd inhibited the growth of tumor cells by 68.14%,55.56%,47.01% and,37.52%.Compared with CTX group,the immune organ index was ameliorated (P<0.01).Compared with the model group,the levels of serum cytokines IFN-γ and TNF-α were increased (P<0.01), while the levels of serum IL-6,ALT,BUN and CRE were decreased (P<0.01).H&E staining of tumor tissue and apoptosis test showed that acid-soluble polysaccharides promoted tumor cell apoptosis.【Conclusion】The acid-soluble polysaccharides from Trametes versicolor (L.) Lloyd have antitumor effect,and may be through the regulation of immune capacity to achieve this effect.
