藏猪源高产纤维素酶菌株的筛选及鉴定

摘要:
【目的】从西藏藏猪粪便中分离可高效降解纤维素的菌株,并对其进行分类和鉴定。【方法】用羧甲基纤维素(CMC)水解圈法对藏猪粪便中的产纤维素酶菌株进行初筛,再用滤纸降解试验、秸秆纤维降解试验和胞外酶活力测定等进行复筛,筛选可高效降解纤维素的细菌。通过观测所获菌株的菌落形态和生理生化特征以及16S rRNA基因序列分析,对菌株进行分类鉴定。【结果】筛选到1株具有较强纤维素降解能力的菌株,经鉴定其是解淀粉芽孢杆菌的一个亚种或变种,将其命名为解淀粉芽孢杆菌(Bacillus amyloliquefaciens)TL106。该菌株的内切葡聚糖酶和β-葡聚糖苷酶活力分别高达86.34和65.54 U/mL。【结论】获得了可高效降解纤维素的菌株TL106,具有较好的应用前景。

Screening and identification of high yield cellulase degrading bacteria strain from Tibetan pigs

Abstract:
【Objective】The aim of this study was to screen and identify cellulase degrading microorganisms from pig dung in Tibet.【Method】A highly efficient cellulose degrading strain was obtained by CMC hydrolysis spot diameter measurement method from Tibetan pig dung and was re screened with disintegration test of filter paper scrip,straw weight lose assay method and of extracellular enzyme activity measurement method.Through the morphological characteristics,physiological and biochemical characteristics and 16S rRNA gene sequence homology,the strain was identified.【Result】A bacterial strain with high cellulose degrading ability was isolated and identified as Bacillus amyloliquefaciens TL106.Its endo 1,4-β-D-glucanase and β-1,4 glucosidase were 86.34 and 65.54 U/mL,respectively.【Conclusion】The obtained TL106 was an efficient cellulose degrading strain with a good application prospect.
