He-Ne激光辐照对灯盏花组培体系的影响

摘要:
【目的】研究He-Ne激光辐照不同时间对灯盏花组织培养的影响,为灯盏花的开发利用提供参考。【方法】以灯盏花的种子、无菌培养苗、叶片、愈伤组织及芽为材料,用He Ne激光辐照不同时间(种子分别为0(CK),0.5,1,5,10 min;叶片分别为0(CK),2,4,6,8 min;愈伤组织分别为0(CK),1,2,3,4,5,6,7,8 min;分化芽分别为0(CK),0.5,1,2,3,6 min;根分别为0(CK),0.5,1,2 min),分析He Ne激光辐照对各组织的影响,筛选最佳辐照时间。【结果】He-Ne激光辐照1 min能够提高灯盏花种子活力;诱导愈伤组织初期,较适宜的辐照时间是2 min,继代后以辐照8 min的愈伤组织生长最好;愈伤组织增殖培养时,辐照4 min效果最佳;诱导芽辐照3 min较适宜;生根培养以辐照2 min较好。【结论】一定量的He-Ne激光能够提高灯盏花种子的萌发率、出愈率及愈伤组织诱导芽和生根效果。

Abstract:
【Objective】Experiment was carried out to study the effects of He-Ne laser on tissue culture system of Erigeron breviscapus and improve E.breviscapus development and utilization.【Method】Seeds,leaves,callus and roots were used as material and different irradiation times were set up.The irradiation times for germination of seed were 0(CK),0.5,1,5,and 10 min,for inducement of leaves were 0(CK),2,4,6,and 8 min,for callus were 0(CK),1,2,3,4,5,6,7,and 8 min,for callus were 0(CK),0.5,1,2,3,and 6 min while for rooting were 0(CK),0.5,1,and 2 min.Then effects of He-Ne laser irradiation were analyzed to select optimum irradiation times.【Result】The suitable irradiation times for improving seed vigor,early to callus,successive culture to callus,during the multiplication culture,bud inducement,and root were 1 min,2 min,8 min,4 min,3 min,and 2 min,respectively.【Conclusion】A certain dose of irradiation was suitable to improve rate of germination and growth of callus,shot,and root.
