| 【目的】研究中药复方灌注液及其中的单味中药提取物对原代培养的奶牛乳腺上皮细胞增殖及总蛋白含量的影响。【方法】采用原代组织块贴壁法获得奶牛乳腺上皮细胞，以质量浓度为5，10，15和20 mg/mL的中药复方灌注液以及紫花地丁、黄芪、泽兰、鸡血藤、诃子提取物，分别作用于对数生长期的奶牛乳腺上皮细胞，确定最适宜中药质量浓度，采用该质量浓度提取物处理对数生长期的乳腺上皮细胞，并分别在处理的1，3和5 d时取样，采用四甲基偶氮唑盐法(MTT法)检测细胞活性，并测定细胞分泌的总蛋白含量。【结果】通过组织块贴壁与胰蛋白酶消化法相结合，得到较纯化的奶牛乳腺上皮细胞。泽兰、鸡血藤、复方灌注液在质量浓度为10 mg/mL时能极显著促进奶牛乳腺上皮细胞增殖(P＜0.01)，泽兰、鸡血藤、诃子、复方灌注液在质量浓度为15 mg/mL时能极显著提高奶牛乳腺上皮细胞总蛋白的含量(P＜0.01)。【结论】适宜质量浓度的中药作用于奶牛乳腺上皮细胞，可以促进细胞的生长及其总蛋白的分泌。 |

| 【Objective】The purpose of this experiment was to study the effects of Chinese herbal compound perfusion and the single herbal extracts on proliferation and total protein content of cow mammary gland epithelial cells.【Method】Purified cow mammary gland epithelial cells were obtained by using the original organization block adherent method,and the Chinese herbal compound perfusion and extracts of major constituents(Herb violae,Radix astragali,Herba lycopi,Herba paederiae,and Fructus chebulae) were prepared.Different levels (5,10,15 and 20 mg/mL) of compound perfusion and extracts were used to treat mammary gland epithelial cells of cows at exponential growth stage.The optimum effective concentration of compound perfusion was determined and was used to treat cow mammary gland epithelial cells.Then,the cell activities were measured by MTT method and TP values were detected at 1,3 and 5 d.【Result】Purified cow mammary epithelial cells were obtained by the original organization block adherent method and trypsin digestion method.After treatment with different Chinese herbal extracts for 6 days,the cell proliferation was significantly promoted by extracts of Herba lycopi and Herba paederiae as well as compound perfusion at 10 mg/mL level (P＜0.01).Total protein content of cow mammary gland epithelial cells was significantly enhanced by extracts of Herba lycopi,Herba paederiae and Fructus chebulae as well as compound perfusion at 15 mg/mL level (P＜0.01).【Conclusion】Chinese herbal perfusion and extracts at suitable concentrations could promote the growth and protein secretion of cow mammary gland epithelial cells. |
