| 【目的】以橙色叶球大白菜(Brassica campestris ssp.pekinensis(Lour.)Olsson)为试材,研究类胡萝卜素的提取条件及其含量和组分的测定方法。【方法】通过单因素试验和L12(43)正交试验,筛选有机溶剂提取类胡萝卜素的最佳提取条件,利用分光光度法对类胡萝卜素的总量进行测定;建立类胡萝卜素成分的RP-HPLC测定体系,并利用RP-HPLC法对类胡萝卜素的主要成分叶黄素、番茄红素、β-胡萝卜素进行分析。【结果】橙色叶球大白菜类胡萝卜素提取的最佳工艺为:以乙醇 丙酮溶液(V(乙醇)∶V(丙酮)=1∶1)为溶剂,料(g)液(mL)比为1∶20,在60 ℃下浸提50 min。利用分光光度法进行类胡萝卜素测定,得出橙色大白菜类胡萝卜素的平均含量为32.847 mg/kg,是普通白菜中的2.467倍。建立了大白菜类胡萝卜素的RP-HPLC测定体系,以甲醇-乙腈溶液(V(甲醇)∶V(乙腈)=55∶45)为流动相,流速1.00 mL/min,柱温30 ℃,检测波长440 nm,进样量10 μL。以叶黄素、番茄红素、β-胡萝卜素为标样,建立了RP-HPLC法测定叶黄素、番茄红素、β-胡萝卜素3种色素的线性方程。相关分析表明,RP-HPLC法与分光光度法测定类胡萝卜素含量的相关系数达到0.973。【结论】利用RP-HPLC法测定橙色大白菜叶球中的类胡萝卜素,其主要成分是叶黄素、番茄红素和β-胡萝卜素等,而普通大白菜中没有番茄红素。 |

| 【Objective】Extraction and determination methods and components of the total carotenoids in the leaf-head of orange-heading Chinese cabbages (Brassica campestris ssp.pekinensis (Lour.) Olsson) were studied.【Method】The optimal conditions for the extraction of carotenoids using organic solvent were screened by single factor analysis and orthogonal analysis,and the total carotenoids of orange heading Chinese cabbages were determined by UV spectmphotometry.RP-HPLC system for carotenoids component determination was established,and used to measure the main ingredients of carotenoids including lutein,lycopene and β-carotene.【Result】The optimal conditions for the carotenoids extraction from orange heading Chinese cabbages were:solvent was ethanol β-acetone(V(ethanol)∶V(acetone)=1∶1),the ratio of solid (g) to liquid (mL) was 1∶20,and extracting temperature was 60 ℃ and extracting time was 50 min.The average content of carotenoids in orange heading Chinese cabbages determined by UV spectmphotometry was 32.847 mg/kg,which was 2.467 times as that of common Chinese cabbages.The conditions of RP-HPLC were:the mobile phase was methanol-acetonitrile(V(methanol)∶V(acetonitrile)=55∶45),the flow rate was 1.00 mL/min,the column temperature was 30 ℃,the UV wavelength was 440 nm,and the injection volume was 10 μL.Using lutein,lycopene,and β-carotene as standard compounds,three linear equations for measuring lutein,lycopene and β-carotene by RP-HPLC method were obtained.The related coefficient between these two methods to determine the carotenoids components in orange heading Chinese cabbages was 0.973.【Conclusion】Based on RP-HPLC method,the main ingredients of carotenoids in orange heading Chinese cabbages were lutein,lycopene,and β-carotene,while common Chinese cabbages contained no lycopene. |
