2-芳基-3,4-二氢异喹啉类抗菌物质对植物种子萌发及幼苗生长的影响

摘要:
【目的】分析2-芳基-3,4-二氢异喹啉类抗菌物质对植物发芽和幼苗生长的影响,为其作为植物抗菌药物应用时的安全性评价提供理论依据。【方法】以溴化2-(2-氯苯基)-3,4-二氢异喹啉为对象,以体积分数0.5%的二甲基亚砜(DMSO)水溶液作为溶剂,配制12.5,25.0,50.0,100.0,200.0 μg/mL 5个质量浓度的供试溶液。将糜子和油菜种子在供试溶液中浸种20 h后,均匀点播于含无离子水的培养皿中,然后在设定条件下进行培养,分别在24,48和72 h测定2种植物种子的发芽率及幼苗根长、茎长、总鲜质量、总干质量和相对含水量,筛选对糜子和油菜种子萌发及生长安全的质量浓度;按照筛选的安全质量浓度分别配制8种溴化二氢异喹啉类化合物的供试溶液,测定并比较各化合物的供试溶液对糜子和油菜种子萌发及幼苗生长的影响。【结果】当溴化2-(2-氯苯基)-3,4-二氢异喹啉的质量浓度≤100.0 μg/mL时,其对糜子和油菜种子的萌发和幼苗生长均安全,因此确定100.0 μg/mL为异喹啉类化合物作为抗菌物质使用的安全质量浓度;在100.0 μg/mL安全质量浓度下,除溴化2-(2-三氟甲基苯基)-3,4-二氢异喹啉溶液对糜子浸泡48 h时的发芽率及油菜根长有轻微抑制作用外,其他7种化合物对2种植物的发芽率及幼苗生长均无显著影响。【结论】当使用质量浓度≤100.0 μg/mL时,大部分异喹啉类化合物对植物安全无害。

Effects of 2-aryl-3,4-dihydroisoquinolin-2-iums on seed germination and seedling growth

Abstract:
【Objective】This study was conducted to analyze the effects of 2-aryl-3,4-dihydroisoquinolin-2-iums as antifungal agent on seed germination and seedling growth so as to improve its safety evaluation as antimicrobial drug for plants.【Method】We selected 2-(2-Chlorophenyl)-3,4-dihydroisoquinolin-2-ium bromide as the research object and five test solutions with different mass concentrations (12.5,25.0,50.0,100.0,and 200.0 μg/mL) were prepared using 0.5% (volume fraction) dimethylsulfoxide (DMSO) aqueous solution as the solvent.The seeds of Panicum miliaceum L.and Brassica campestris L.were soaked in the test solutions for 20 h,and then evenly sowed in a Petri dish containing deionized water.At 24,48 and 72 h,the germination ratio,and the average root length,stem length,gross fresh weight,gross dry weight and relative water content of seedlings in each group were determined,respectively.Then the safe concentration for seed germination and seedling growth of two plants were determined.8 test solutions of dihydroisoquinolin-2-ium bromide under the determined safe mass concentration were prepared and their effects on seed germination and seedling growth of P.miliaceum L.and B.campestris L.were compared.【Result】2-(2-Chlorophenyl)-3,4-dihydroisoquinolin-2-ium bromide was safe to the seed germination and seedling growth of the two plants when its mass concentration was equal to or lower than 100.0 μg/mL.Thus,100.0 μg/mL was regarded as the safe mass concentration of Isoquinoline compounds as antifungal agents.With the concentration of 100.0 μg/mL,2-(2-Trifluoromethylphenyl)-3,4-dihydroisoquinolin-2-ium bromide slightly inhibited the germination of P.miliaceum L.after being soaked for 48 h and the average root length of B.campestris L. seedling,while all other compounds had no significant influence on seed germination and seedling growth of the two plants.【Conclusion】When the concentration was ≤100.0 μg/mL,most of the tested Isoquinoline compounds were safe to plants.
