| 【目的】探索2个德国鸢尾品种金娃娃、黑骑士的组织培养条件,为2种鸢尾的种苗产业化和推广应用奠定基础。【方法】以金娃娃、黑骑士2种鸢尾的幼嫩花葶为材料,研究不同外植体、消毒方法和培养基对2种鸢尾不定芽诱导、增殖及生根的影响。【结果】2种鸢尾的花茎节能直接诱导出不定芽,故其为组织培养的最佳外植体;金娃娃外植体以75%(体积分数)乙醇浸摇3 s、0.1%(质量分数)HgCl2消毒6 min的处理污染率和死亡率均较低;黑骑士则以75%乙醇浸摇5 s、0.1% HgCl2消毒8 min的效果较好。初代培养中,MS+2.0 mg/L 6-BA+0.1 mg/L NAA是2种鸢尾不定芽诱导的最佳培养基,在该培养基中,金娃娃和黑骑士的不定芽诱导率均最高,分别为86.4%和73.5%。继代培养中,MS+4.0 mg/L 6-BA+0.2 mg/L NAA是不定芽增殖的最佳培养基,在该培养基中,金娃娃和黑骑士的增殖系数最高,分别为12.71和8.64。2种鸢尾不定芽生根培养时以培养基MS+0.1 mg/L 6-BA+2.0 mg/L NAA效果最好。【结论】建立了2个德国鸢尾品种金娃娃、黑骑士的最佳组织培养体系,在该体系下不定芽诱导率、增殖率高,生根情况良好。 |

| Abstract: |
| 【Objective】This study investigated the tissue culture conditions for Iris germanica‘Gold Boy’ and ‘Black Knight’,to improve the seedling industrialization and application.【Method】Using young scapes as materials,effects of different explants,disinfection methods and culture media on the induction of adventitious buds, proliferation of adventitious buds,rooting of buds of two irises were investigated. 【Result】Adventitious buds were only induced from the tubers of flower stalks.Soaking for 3 seconds in 75% (volume fraction) alcohol and for 6 min in 0.1% (mass fraction) HgCl2 forIris germanica‘Gold Boy’,while 5 seconds in 75% alcohol,and 8 min in 0.1% HgCl2 gave the lowest contamination rate and death rate for ‘Black Knight’.The optimal medium of primary culture for the induction of adventitious buds was MS+2.0 mg/L 6-BA+0.1 mg/L NAA with induction rates of 86.4% and 73.5% for ‘Gold Boy’ and ‘Black Knight’,respectively.As for secondary culture,MS+4.0 mg/L 6 BA+0.2 mg/L NAA was the optimal medium with the highest proliferation coefficients of 12.71 and 8.64 for ‘Gold Boy’ and ‘Black Knight’,respectively.MS+0.1 mg/L 6-BA+2.0 mg/L NAA was the optimal medium for rooting.【Conclusion】Optimal culture system for two Iris germanica cultivars ‘Gold Boy’ and ‘Black Knight’ was established with high induction and proliferation rates of adventitious buds and good rooting growth. |
