| 【目的】探讨影响川滇高山栎体胚诱导的关键因素,为建立川滇高山栎体胚再生植株体系奠定基础。【方法】以川滇高山栎未成熟合子胚为外植体,研究基本培养基、植物生长调节剂配比、采样时期、抗氧化剂以及培养条件(光培养或暗培养)对其体胚诱导的影响。【结果】川滇高山栎未成熟合子胚在MS、1/2MS、B-5和WPM基本培养基上均有体胚发生,其中MS基本培养基诱导效果较好,诱导率达60.0%,显著高于其他培养基,且愈伤组织量最多;以MS为基本培养基,同时添加1 mg/L 6-BA和0.5 mg/L 2,4-D,诱导率达83.3%,显著高于二者单独添加及其他组合处理;光照强度1 000~2 000 lx,每天光照12 h的光培养比暗培养更有利于体胚的发生;在林芝地区最佳采样时期为08-10左右;培养基中添加1.0 g/L PVP可有效减轻褐变的发生。【结论】初步建立了川滇高山栎体胚诱导的培养体系。 |

| 【Objective】This study investigated the factors that influence the induction of somatic embryogenesis in Quercus aquifolioides Rehd.et Wils.【Method】Using immature zygotic embryos of aquifolioides as explants,effects of basic medium,hormones,concentrations of antioxidants medium,and culturing conditions on the induction of somatic embryogenesis were investigated.【Result】Somatic embryos were induced in all four tested basic medium including MS,1/2 MS,B-5,and WPM.MS medium had the best induction rate of 60.0% with the maximum amount of callus.High induction rate of 83.3% was achieved in MS medium supplemented with 1 mg/L 6-BA and 0.5 mg/L 2,4-D,which was significantly higher than all other treatments.Light intensity of 1 000-2 000 lx for 12 h per day was more conducive to the occurrence of somatic embryogenesis than dark culture.Optimal sampling period was around August 10th and medium supplemented with 1.0 g/L PVP effectively reduced browning.【Conclusion】A culture system with optimal culture conditions for induction of somatic embryogenesis of Quercus aquifolioides Rehd.et Wils was established. |
