甲氰菊酯降解菌ZH-3的分离鉴定及降解特性

摘要:
【目的】分离筛选甲氰菊酯的高效降解菌株,为拟除虫菊酯类杀虫剂果蔬残留危害的综合治理提供候选生物制剂。【方法】采用基础盐培养基,从农药厂废水排放口的污泥中筛选降解菌,以甲氰菊酯为唯一碳源进行摇瓶培养复筛,以降解率为评价标准,确定高效降解菌株,根据形态、生理生化特征和16S rDNA序列同源性鉴定其种属,高效液相色谱法研究其降解特性。【结果】分离得到甲氰菊酯高效降解菌株ZH-3,初步鉴定为蜡状芽孢杆菌 (Bacillus cereus )。菌株ZH-3能有效降解25~300 mg/L的甲氰菊酯,在1%接种量、30 ℃、pH 8.0、160 r/min条件下,3 d内对50 mg/L甲氰菊酯的降解率为85.3%。【结论】菌株ZH-3对菊酯类农药的降解作用谱广,对甲氰菊酯、高效氯氰菊酯、氯氟氰菊酯、氯氰菊酯及溴氰菊酯等均具有较高的生物降解作用。

Isolation,identification and degradation characteristics of a fenpropathrin degrading bacterium ZH-3
Abstract:
【Objective】The study was conducted to isolate-effective bacteria strains for biodegrading fenpropathrin,to provide biological agents for the comprehensive management of fruits and vegetables against residual pyrethroid pesticides.【Method】 The degrading bacteria strains were isolated in mineral salt medium from a waste water outlet of a pesticide plant, and then screened in the media of fenpropathrin as only carbohydrate.The effective degrading bacteria strains were determined according to the degrading rate and preliminarily identified based on its morphology,physiology and homologic analysis of 16S rDNA sequence.【Result】 Fenpropathrin-degrading bacterium strain ZH-3 was isolated and preliminarily identified as Bacillus cereus.ZH-3 could effectively degrade fenpropathrin ranging of 25-300 mg/L,and degrade 85.3% of 50 mg/L fenpropathrin in 3 d,under the conditions of 1% inoculum,30 ℃,pH 8 and 160 r/min.【Conclution】 ZH-3 has a broad spectrum of degradation and great high biodegradation efficiency towards pyrethroid pesticides,such as fenpropathrin,cypermethrin,cyhalothrin,deltamethrin and cypermethrin.
