| 【目的】探讨泌乳期小鼠下丘脑-垂体-卵巢轴中催乳素释放肽(PrRP)及其受体(PrRP-R) mRNA的表达规律,为PrRP生理功能的确定奠定基础。【方法】选用泌乳6,12,18 d小鼠,取下丘脑、垂体、卵巢组织,利用半定量PCR方法测定下丘脑、垂体、卵巢中PrRP、PrRP-R mRNA的表达水平;同时利用放射免疫法(RIA)测定血浆的孕酮(P)、雌二醇(E2)和催乳素(PRL)水平,通过双侧相关分析,探究血浆P、E2、PRL水平与下丘脑-垂体-卵巢轴PrRP、PrRP-R mRNA表达量的相关关系。【结果】泌乳期小鼠下丘脑、垂体、卵巢中均有PrRP、PrRP-R mRNA的表达,其中卵巢的PrRP mRNA表达水平最高。泌乳期小鼠血浆E2水平与卵巢PrRP mRNA的表达水平呈显著负相关,而卵巢的PrRP mRNA表达水平与垂体的PrRP-R mRNA表达水平呈显著正相关。【结论】泌乳期小鼠卵巢PrRP可能通过垂体间接抑制E2的分泌,进而调节相关泌乳活动。 |

| 【Objective】The research was made to investigate the function and expression of PrRP and PrRP-R mRNA in hypothalamus-pituitary-vary axis during lactation.【Method】Hypothalamus,pituitary and ovary were sampled on 6,12 and 18 d of lactation respectively for determination of PrRP and PrRP-R mRNA expression levels in these tissues by semi-RT-PCR.Simultaneously,plasma concentration of progestin(P),estrogen(E2) and prolactin(PRL) were measured by RIA.Corelation between PrRP,PrRP-R mRNA and plasma P,E2 and PRL were analyzed to investigate the relations among them.【Result】During lactation,hypothalamus,pituitary and ovary all showed PrRP and PrRP-R mRNA expression.Ovary had the highest PrRP mRNA level.Plasma E2 had significant negative correlation with ovary PrRP mRNA and ovary PrRP mRNA had significant positive correlation with pituitary PrRP-R mRNA.【Conclusion】Ovary PrRP might indirectly inhibit the secretion of E2 by feedback on pituitary. |
