杆状病毒表面展示系统及其应用研究进展

摘要:
杆状病毒表面展示系统是近年来发展起来的一种新的真核生物表面展示系统.通过与杆状病毒囊膜蛋白gp64融合表达或与特异性的锚定部位结合,可以将外源肽或蛋白质展示在杆状病毒表面, 形成所谓的杆状病毒"假病毒",从而筛选出目的蛋白或活性肽.杆状病毒表面展示系统可用来展示需要磷酸化、糖基化、甲基化、乙酰化、羟基化、二硫键异构化等翻译后修饰,才能表现功能活性的复杂真核蛋白,以及用于构建多肽文库和抗体库、新型疫苗研制、基因治疗等方面.文章对杆状病毒表面展示系统的原理、特点和应用方面进行了综述,并对其发展前景进行了展望.

The advance in Baculovirus surface display system and its application
Abstract:
〗Baculovirus surface display system is a new eukaryotic display technology developed in recent years.Through fused to the entire membrane protein gp64 of baculovirus or combined with special membrane anchor domain,exotic peptides and proteins could be expressed on the surface of baculovirus,then "pseudotyped baculovirus" could be produced and aim proteins and bioactive peptides could be screened out.Baculovirus surface display system can be used to display complex eukaryotic proteins requiring post translational processing including hosphorylation,methylate,acetylize,hydroxylation,disulfide linkage isomerizate efficient folding for functional activity.In addition,Baculovirus surface display system can be used to construct polypeptide library,antibody library,new vaccine manufacture,gene therapy et al.In this paper,we mainly described the principles,characteristics,applications and prospects of the baculovirus surface display system.
