| 为探讨IGF-1基因对北京鸭胸肌发育的影响,试验克隆了北京鸭IGF-1 cDNA的部分序列,分析了其核苷酸及编码的氨基酸序列与其他物种的同源性,并检测了IGF-1 mRNA在不同日龄北京鸭胸肌中表达的发育性变化.结果表明,克隆的鸭IGF-1 cDNA部分序列长507 bp,编码168个氨基酸,核苷酸序列与鸡、家鸭、鹌鹑、火鸡、鸵鸟、人和猪的同源性分别为98%,99%,97%,98%,96%,84%和82%,编码的氨基酸序列与家鸭、鸡、火鸡、鹌鹑、人、挪威鼠和猪的同源性分别为99%,98%,97%,97%,84%,79%和84%.表明北京鸭与家鸭的亲缘关系最近,与鸡、鹌鹑、火鸡、鸵鸟也有较近的亲缘关系;北京鸭胸肌中IGF-1 mRNA表达丰度在7,14,21日龄间和28,35,42日龄间的差异均不显著(P>0.05),但28,35,42日龄显著高于7,14,21日龄(P<0.05),说明在北京鸭胸肌发育较快的时期,IGF-1 mRNA表达量也较高,IGF-1 mRNA的表达量与胸肌的发育呈正相关. |

| Molecular cloning of IGF-1 partial cDNA and the development change of expression in the Beijing duck''''s breast muscle |

| In order to research the effect of IGF-1 to the breast muscle development of Beijing duck,we cloned the partial cDNA sequence of duck IGF-1 gene and analyzed the identity of nucleotide sequence and deduced amino acid sequence between Beijing duck and other species.At the same time,the development of change of IGF-1 expression in the Beijing duck's breast muscle was discussed.The results showed that 507 bp cDNA of Beijing duck IGF-I gene were cloned.The duck IGF-I cDNA coded for a predicted protein of 161 amino acids.Cloning Beijing duck IGF-I cDNA partial sequence has an overall similarity with a comparable region of chicken (98%),domestic duck (99% ),coturnix (97%),turkey (98% ),ostrich (96%),human(84%) and pig (82%).It was found to have 99%,98%,97%,97%,84%,79% and 84% sequence similarity at the predicted amino acid sequence with domestic duck ,chicken,turkey,quail,human,Norway rat and pig respectively.The results showed that the Beijing duck is most closely related to that of the domestic duck,and more closely related to that of the chicken,ostrich,quail,and turkey than to that of the other species.IGF-1 mRNA expression abundance in the Beijing duck's breast muscle was not significantly different at the age of 7 d,14 d,21 d and 28 d,35 d,42 d,respectively.But,IGF-1 mRNA expression abundance in the Beijing duck's breast muscle at 28,35 and 42 d of age was higher than that of at 7,14 and 21 d of age.Beijing duck's breast muscle IGF-1 mRNA levels increased following the fast development of breast muscle.It existed the positive correlation between IGF-1 expression and law of breast muscle development. |
