野生黄木香的离体培养及其再生体系研究

摘要:
以野生黄木香茎段为外植体,探讨了不同生长调节物质对其不定芽诱导、增殖和生根的影响,并建立了野生黄木香的高效再生体系。结果表明,利于野生黄木香分化的诱导培养基为MS+AgNO3 3.0 mg/L+6-BA 1.5 mg/L+NAA 0.5 mg/L+琼脂6.5 g/L+蔗糖30 g/L;以MS+AgNO3 3.0 mg/L+6-BA 1.5 mg/L+NAA 0.05 mg/L+琼脂6.5 g/L+蔗糖30 g/L为增殖培养基时的增殖效果最好,增殖倍数为5.5;黄木香高效的再生体系为1/2 MS+AgNO3 4.5 mg/L+IBA 0.2 mg/L+质量分数0.2%活性炭+琼脂4.0 g/L+蔗糖20 g/L,其生根率可达95 %。

Culturing of wild Rosa banksiae f.lutescecs Vots.in vitro and plant regeneration system
Abstract:
By taking wild Rosa banksiae f.lutescecs Vots.as explants,the influence of different growth regulating substances on adventitious buds's induction,multiplication and rooting was studied,and the regeneration system of high efficiency on Rosa banksiae f.lutescecs Vots.was established.The results suggested that the best medium for induction was MS+AgNO3 3.0 mg/L+6-BA 1.5 mg/L+NAA 0.5 mg/L+agar 6.5 g/L+sucrose 30 g/L;the best medium for the multiplication was MS+AgNO3 3.0 mg/L+6-BA 1.5 mg/L+NAA 0.05 mg/L+agar 6.5 g/L+sucrose 30 g/L,and the multiplication multiple was 5.5;the regeneration system of high efficiency on Rosa banksiae f.lutescecs Vots.was 1/2 MS+AgNO3 4.5 mg/L+IBA 0.2 mg/L+0.2% activated carbon+agar 4.0 g/L+sucrose 20 g/L,and the rooting rate was 95%.
