| 对用发酵法从鲁氏毛霉中制备、分离、提取甲壳素和壳聚糖的方法和工艺条件进行了系统研究。结果表明:(1)鲁氏毛霉较佳培养基组成为果糖20 g/L,牛肉膏15 g/L,K2HPO4 0.75 g/L,MgSO4 1.80 g/L,N-乙酰葡萄糖胺1.54 g/L;其较优发酵条件为温度28 ℃,周期48 h,pH5.0,摇瓶装液量200 mL/L。(2)从菌丝体中提取甲壳素和壳聚糖的工艺条件:除蛋白质为氢氧化钠20 g/L,温度115 ℃,时间60 min;酸处理为盐酸30 g/L,温度80 ℃,时间180 min。(3)采用较优发酵提取条件后,甲壳素产量可达1.328 g/L,壳聚糖产量达0.672 g/L,壳聚糖占菌粉干重的7%。 |

| A study on the preparing of chitin and chitosan by fermentation with Mucor rouxianus |

| It was systematically studied on the methods and technology of preparing,isolating chitin and chtosan by fermentation with Mucor rouxianus.As the experiment indicating:(1) The optimal composition of fermentation medium for Mucor rouxianus was:fructose 20 g/L,beef extract 15 g/L,K2HPO4 0.75 g/L,MgSO4 1.80 g/L,acetyl glucosamine 1.54 g/L;The optimal condition of fermentation was:temperature 28 ℃,time 48 h,pH 5.0,amount of medium per flask 200 mL/L.(2) The conditions of isolating chitin and chitosan from the mycelium were:① isolating protein:NaOH 20 g/L,temperature 115 ℃,time 1 h;② acid treatment:HCl 30 g/L,temperature 80 ℃,time 3 h.(3) Under the optimal conditions of isolation:yield of Mucor rouxianus was:chitin 1.328 g/L,chitosan 0.672 g/L,percent of chitosan in dried mycelium was 7%. |
