| 应用酶活性测定和聚丙烯酰胺浓度梯度凝胶电泳技术,研究了3种不同浓度的重金属胁迫对不同类型水稻萌发种子中α,β-淀粉酶同功酶活性的影响。结果表明:(1)0.05 mmol/L的Cu2+ ,Cd2+,Hg2+胁迫使水稻萌发种子中α-淀粉酶活性有所增高 ,而>0.05 mmol/L的 Cu2+,Cd2+,Hg2+胁迫则导致水稻各品种萌发种子中β-淀粉酶活性显著降低。 (2 ) Cu2+,Cd2+,Hg2+胁迫使武育粳萌发种子中β-淀粉酶活性随重金属浓度的增高呈先上升后下降的趋势 ,并显著抑制(尤其是 Hg2+)扬稻6号和汕优818的β-淀粉酶活性。(3) 0.05 mmol/L的Cu2+,Cd2+,Hg2+胁迫能明显诱导新的淀粉酶同功酶的产生 ,>0.05 mmol/L的Cu2+,Cd2+,Hg2+则抑制一些同功酶的正常表达 |

| Effect of heavy metal stress on the amylase activity in germinating rice seeds |

| In this paper,the α,β-amylase activity measuring methods and polyacrylamide concentration gradient gel electrophoresis technique were used to study the effect of heavy metal stress on amylase activity in germinating rice seeds.The results showed:(1) 0.05 mmol/L Cu2+,Cd2+ ,Hg2+ stress caused α-amylase activity in germinating Wuyujing,Yangdao 6,Xianyou 818 seeds to increase obviously.However,the higher concentration of Cu2+,Cd2+ ,Hg2+ stress caused α-amylase activity to decrease markedly,and the inhibited degree of α-amylase activity enhanced with the Cu2+,Cd2+ ,Hg2+ concentration decreasing.(2) With the concentration of Cu2+,Cd2+ ,Hg2+ increasing,the β-amylase activity in germinating Wuyujing seeds increased and then decreased.The activity of β-amylase in germinating Yangdao 6 and Xianyou 818 seeds was inhibited by all Cu2+,Cd2+ ,Hg2+ treatments (especially,the Hg2+).(3) Although>0.05 mmol/L Cu2+,Cd2+ ,Hg2+ stress inhibited the expression of some amylase isozymes which were expressed in normal condition(such as in control),0.05 mmol/L Cu2+,Cd2+ ,Hg2+ stress induced the expression of some amylase isozymes significantly. |
