波形纤维蛋白在前体脂肪细胞分化中的表达

摘要:
采用免疫组化(HIGH-SABC)法对前体脂肪细胞分化中波形纤维蛋白的表达进行了定性和定量检测,并结合计算机图像分析系统对细胞形态参数值(细胞总面积、胞核面积、细胞周长、等效直径及胞核质比)进行了分析。结果表明:(1)波形纤维蛋白参与前体脂肪细胞分化的全过程,而且其表达具有明显的时间依赖性---随分化的进行呈下降的趋势,染色模式为细胞质;(2)在前体脂肪细胞积聚滴向脂肪细胞分化的过程中,细胞总面积、胞核面积、细胞周长和等效直径总的呈上升趋势,胞核质比呈下降倾向。

Expression of vimentin during the differentiation of rat preadipocyte
Abstract:
The effects of differentiation of rat preadipocytes into adipocytes on vimentin protein expression was determined by immunohistochemistry,using a monoclonal anti-rat vimentin antibody,on the morphologic parameters (whole cell area,nucleus area,cell perimeter,equivalent diameter,and N:C ratio) of the cells oberserved by cell image analysis.The results demonstrated (1):the immunophenotype of IF was vimentin+,staining intensity varied as cultured rat preadipocytes became differentiated,usually showing more intense staining for the vimentin systems at the preadipocytes during early differentiation compared to the rounded lipid filled adiopocytes undergoing changes in morphology.Downregulation of vimentin was required for adipose conversion and expression of vimentin showed a time-dependence;(2) the differentiation of growing preadipocytes into adipocytes led to a strong increase in the cell equivalent diameter,perimeter,surface area and volume and a reduction of N:C ratio.
