德国骑乘马线粒体DNA的限制性酶分析

摘要:
通过5种限制性内切酶获得了13匹德国骑乘马线粒体DNA(mtDNA)的切割图谱。结果发现,限制性消化产生了1(EcoRⅠ), 3(BglⅡ), 4(BamHⅠ)或5个(HindⅢ和BamHⅠ)限制性片断,并确定了所有片断的分子大小。mtDNA总的大小平均为(16. 48±0. 10) kb.除BamHⅠ外,mtDNA的限制性切点对于所有的个体是完全一致的。用BamHⅠ检测到mtDNA的多态性,通过带谱分析推断,德国骑乘马的B型m tDNA是由A型mtDNA通过突变产生1个新的切割位点进化而来。

Restriction Endonuclease Analysis of Mitochondrial DNA of German Saddle-Horse
Abstract:
13 German saddle-horses cleavage patterns of mtDNA from 5 restriction endonucleases (BamHⅠ,HindⅢ,EcoRⅠ,EcoRV and BglⅡ) were obtained. The restriction digests yielded one (EcoRⅤ), 3 (BglⅡ), 4 (BamHⅠ) or 5 (HindⅢ and BamHⅠ) fragments and all fragment sizeswere given.The mean total size of mtDNA was calculated to be (16. 48±0. 10) kilobases.The restriction cleavage sites of mtDNA for all individuals except BamHⅠwere completely identical.A polymorphism of mtDNA was detected with BamHⅠ.The kind of polymorphism let us assume that B-type mtDNA of this horse had evolved from the A-type mtDNA by acquiring an additional cleavage site.
