家兔精子体外获能与体外受精试验

摘要:
本试验目的在于建立一种简便而有效的系统,能使家兔射出的精子完成体外获能,并获得体外受精、早期卵裂和正常产仔。精子体外获能采用三种方法:A.HIS(15min)+DM(1~2h);B.HIS(15min)+DM(11~12h);C.m-HIS(15min)+m-DM(2~4h)。经上述方法处理的精子与512枚输卵管卵母细胞进行体外受精和体外培养的结果表明,A组的受精和早期卵裂发育延迟;B组的受精卵大都停留在原核期;C组的受精和早期卵裂发育正常。将C组60枚2-4细胞移植至9只受体兔,5只妊娠,其中2只产4只试管仔兔,另外3只获得12个胎儿。

Abstract:
The purpose of this research was to establish an available simplified method that would enable ejaculated rabbit sperm to coplete in vitro capacitation and obtain in vitro fertilization, early cleavage and normal offspring of rabbit ova. In vitro capacitation of sperm was carried out by ; using three methods. A. in high,, ;iprue strength solution followed by prein-cubation in defined medium for 1-2 hours) B.in HIS followed by preincu-bation in DM for 11-12 hours C. in m-HIS ( 10 mL m-DM + 34 mg NaCl) followed by preincubation in m-DM (100 mL DM + 84mg NaHCO3) for 2-4 hours. Five hundred and twelve oocytes were cocultured in DM or m-DM with above three kinds of capacitated sperms.The fertilization and early cleavage were retarded in group Aj the fertilization ova almost stayed in pronucleus period in group Bj good result of fertilization and early cleavage were obtained in group C. Sixty 2-4 cell embryos from group C were transferred into nine recipients, five of which became pregnant. Four embryos were carried successfully to deliver normal offspring by two reci-i. i , pients.. Laparotomies were, performed in-three recipients to assess Implantationabout two weeks after embryo transfer, and twelve emhryos were implanted.
