猪精液冷冻保存的初步研究

摘要:
采用液氮熏蒸法制作猪冻精颗粒,对稀释液及冷冻保护剂进行优化筛选。结果表明,在5种稀释液中,5号稀释液优于其他4种(P<0.05);甘油的冷冻保护性能优于乙二醇和二甲基亚砜(P<0.01),其适宜浓度为2 mL/L;稀释液中添加安钠咖可有效延长精子的冻后存活时间(P<0.01)。猪精液冷冻效果的关键因素是冷冻温度、冷冻速度和解冻温度,通过对不同温度情况的研究,初步制定了猪精液冷冻温度曲线。

Study on the technique of boar semen freezing
Abstract:
The diluents,the cryoprotectant concentration and freezen curve used to produce frozen boar semen pellets were studied.The results showed that the diluent 5 is the best one (P<0.05)in five groups,and glycerol is better than glycol or DMSO as the cryoprotectant (P<0.01).When the glycerol concentration is 2 percent,the sperm motility of frozen semen pellets is the best in all treatment groups.Supplying CNB(Caffeine and Natrium Benzoate) in the diluent can increase the thawy sperm survival time and motility effectively (P<0.01).The freezing temperiture,freezing rate and thawy temperiture are the keys which influence the effect on boar freezing semen.The freezing curve of boar semen is established base on studing the different temperature.
