蒙古羊、兰州大尾羊和哈萨克羊随机扩增多态DNA分析

摘要:
从24个随机引物中筛选出5个引物,对蒙古羊、兰州大尾羊和哈萨克羊3个绵羊品种共计89个个体进行了随机扩增多态DNA分析,根据扩增片段计算了蒙古羊、兰州大尾羊和哈萨克羊扩增DNA指纹条带的平均带纹数分别为5.32,5.28和5.78;平均带纹相似率分别为0.59,0.60和0.62;平均杂合度分别为0.75,0.78和0.77。用Nei氏片段共享度公式计算了3个群体的遗传距离,构建了其关系图,聚类分析结果表明,蒙古羊与兰州大尾羊的遗传距离最小(0.034),彼此亲缘关系较近。相对而言,兰州大尾羊与哈萨克羊的遗传距离最大(0.104),彼此亲缘关系较远。

Study on random amplified polymorphic DNA fingerprints in Mongolian,Lanzhou big tail and Kazak sheep breeds
Abstract:
Random amplified polymorphic DNA (RAPD) analysis was used to detect the genetic polymorphisms of 89 individuals of three sheep breeds from Mongolian,Lanzhou big tail and Kazak sheep breeds in China by 5 random primers.The average band sharing rates and probability for a band to be present in the heterozygous state of the three sheep populations were calculated to be 0.59 and 0.75 in Mongolian sheep,0.60 and 0.78 in Lanzhou big tail sheep and 0.62 and 0.77 in Kazak sheep,respectively.The genetic distance was obtained by BSR.Dendrogram of three sheep breeds was constructed.The results indicated that the relatively closer genetic relationship was between the Mongolian sheep and Lanzhou big tail sheep.Comparatively,the relationship between Lanzhou big tail sheep and Kazak sheep was rather far.
