犬细小病毒的分离与鉴定

摘要:
从临床表现体温升高、呕吐、血样腹泻、脱水等疑似细小病毒(Canine Parvovirus,CPV)感染的病犬中,采取粪样8份。应用狗肾传代细胞(MDCK)增毒,其中6号病料(CPV6)在MDCK细胞上产生脱落、变形、游离等细胞病变,且细胞感染性试验发现,CPV6对MDCK的感染性强于对猫肾传代细胞(CRFK)的感染性;核酸型试验证明,CPV6毒株的代谢可被5 溴脱氧尿核苷(FUDR)所抑制,其核酸属于DNA型;所分离的病毒培养物能凝集猪、猴、马、猫的红细胞,凝集效价达26~28,并能被已知犬细小病毒阳性血清所抑制,但不能凝集牛、犬、绵羊、兔、小鼠和鸡的红细胞;电镜观察病毒粒子外观呈圆形或六边形,直径约20~23 nm;该病毒耐酸、耐热、耐乙醚;动物致病性试验表明,经口服1 mL,试验组幼犬第7天发病,采集病犬粪样做HA试验为阳性反应,其凝集猪红细胞的特性能被犬细小病毒阳性血清所抑制。

Isolation and identification of canine parvovirus
Abstract:
Eight feces specimens of dogs which have seemingly been infected by canine parvovirus (CPV) according to the clinical signs such as fevering,vomiting,bloody diarrhea and dehydration etc.were collected and proliferated by using MDCK.It was found that the No.6 specimen (CPV6) caused MDCK to have given some pathological changes of desquamation,metamorphism and dissociation etc.The infection ability of CPV6 to MDCK was stronger than that of CPV6 to CRFK by cellular infection test.The metabolism of CPV6 can be inhibited by FUDR and it was a kind of DNA virus.It was also found that CPV6 culturing medium can agglutinate the red blood cells of pigs,monkeys,horses and cats.The diluter of HA is in the range of 26 to 28.And the red blood cells of bovines,canines,sheep,rabbits,mice and chooks cannot be agglutinated by the CPV6 culturing medium.The appearance of CPV is rotundity or hexagon under transmission electron microscope (TEM) and its diameter is about 20-23 nm.It can withstand acid,heat and ether.The pups in the test group embodied CPV infection on the 7th day after they have taken 1 mL of CPV6 culturing medium in the test of onsogeneticity.The feces specimen of those sick pups was positive in the test of HA,and their agglutinating pig red blood cells can be inhibited in the test of HI.
