铝胁迫下八仙花HmVALT基因表达的差异

摘要:
【目的】了解八仙花HmVALT基因表达与耐铝能力的关系,揭示八仙花耐铝机理。【方法】以耐铝的八仙花品种H.macrophylla cv ‘Coerulea’为材料,扩增获得HmVALT基因片段,利用半定量RT-PCR方法,分析在0,50,100,200,400,600 μmol/L AlCl3胁迫下,八仙花根和叶中HmVALT基因的表达情况。【结果】各处理HmVALT基因在八仙花根和叶部均有表达,且根部的表达量高于叶部;铝胁迫浓度为400 μmol/L时八仙花根和叶中HmVALT基因表达量最高,随后降低。【结论】HmVALT基因的表达与八仙花耐铝性密切相关。

Difference in HmVALT gene expression of Hydrangea macrophylla under different aluminum concentrations

Abstract:
【Objective】This experiment investigated the relationship between expression of HmVALT gene and aluminum resistance of Hydrangea macrophylla to provide basis for study on aluminum tolerant mechanism of H.macrophylla.【Method】The HmVALT gene segment was obtained from H.macrophylla cv‘Coerulea’ via RT-PCR.Semi-quantitative RT-PCR was used to analyze the expression variation of HmVALT gene under aluminum stress with different concentrations of 0, 50,100,200,400,and 600 μmol/L. 【Result】The expression of HmVALT gene was found in all treatments,and the expression level in root was higher than that in leaf.The highest expression was obtained with AlCl3 concentration of 400 μmol/L,and then it decreased.【Conclusion】The expression of HmVALT gene was closely related to aluminum tolerance of H.macrophylla.
