不同碳源和氮源对4种深色有隔内生真菌生长的影响

摘要:
【目的】明确不同碳源及氮源对4种深色有隔内生真菌(Dark septate endophytes,DSE)生长的影响,确定4种DSE生长的最适碳源和氮源,为深色有隔内生真菌的研究和应用奠定基础。【方法】以沙门外瓶柄霉(Exophiala salmonis)、甘瓶霉(Phialophora mustea)、菊异茎点霉(Paraphoma chrysanthemicola)和枝状枝孢菌(Cladosporium cladosporioides)4种深色有隔内生真菌为材料,在固体和液体条件下培养,研究不同碳源(葡萄糖、蔗糖、麦芽糖、甘露醇、果糖、乳糖、可溶性淀粉和山梨醇)和氮源(磷酸氢二铵、草酸铵、硫酸铵、氯化铵、尿素、硝酸铵、硝酸钠、酸水解酪蛋白、胰蛋白胨和蛋白胨)对其菌丝平均生长速度和菌丝干质量的影响。【结果】固体培养条件下,沙门外瓶柄霉、甘瓶霉和菊异茎点霉的最佳碳源均是葡萄糖,枝状枝孢菌的最佳碳源是乳糖;沙门外瓶柄霉、甘瓶霉和枝状枝孢菌的最佳氮源均是硝酸钠,菊异茎点霉的最佳氮源是胰蛋白胨。液体培养条件下,甘瓶霉、菊异茎点霉和枝状枝孢菌的最佳碳源均是可溶性淀粉,沙门外瓶柄霉的最佳碳源是葡萄糖;沙门外瓶柄霉、甘瓶霉、菊异茎点霉和枝状枝孢菌的最佳氮源分别是磷酸氢二铵、草酸铵、蛋白胨和硝酸铵。【结论】初步确定了4种深色有隔内生真菌生长的最佳碳源和氮源。

Effects of different carbon and nitrogen sources on growth of 4 dark septate endophytes

Abstract:
【Objective】The objective of this research was to understand the effect of different carbon and nitrogen sources on the growth of 4 dark septate endophytes,and find the optimum carbon and nitrogen sources.The results will provide basis for further study and application of dark septate endophytes.【Method】Exophiala salmonis,Phialophora mustea,Paraphoma chrysanthemicola and Cladosporium cladosporioides were cultured in both solid and liquid medium to study the effects of different carbon (glucose,sucrose,maltose,mannitol,fructose,lactose,soluble starch,and sorbitol) and nitrogen (diammonium phosphate,ammonium oxalate,ammonium sulfate,ammonium chloride,urea,ammonium nitrate,sodium nitrate,acid hydrolyzed casein,tryptone,and peptone) sources on average growth rate and biomass.【Result】Under solid culture condition,the optimum carbon source was glucose for all endophytes except C.cladosporioides,which showed the highest growth on lactose.The optimum nitrogen source of P.chrysanthemicola was tryptone,while the optimum nitrogen source of all others was sodium nitrate.Under submerged culture condition,the optimum carbon source of E.salmonis was glucose,while the optimum carbon source of all others was soluble starch.The optimum nitrogen source of E.salmonis,P.mustea,P.chrysanthemicola and C.cladosporioides was diammonium phosphate,ammonium oxalate,peptone,and ammonium nitrate,respectively.【Conclusion】The nutritional cultivating conditions of 4 dark septate endophytes were determined.
