4种桑葚花色苷的超声提取及其抗氧化能力比较

摘要:
【目的】超声提取黑果桑、白果桑、野生蒙桑和栽培蒙桑桑葚花色苷,分析比较其花色苷含量及抗氧化能力,为吉林地区花色苷含量高、抗氧化能力强的桑种的选育提供参考。【方法】以桑葚冻干粉为试样,在单因素试验的基础上利用响应面分析法确定4种桑葚花色苷的最佳超声提取条件;采用AB8大孔树脂对花色苷进行纯化,测定4种桑葚花色苷的氧自由基清除能力和还原能力,并与同等质量浓度的VC进行比较。【结果】响应面法优化得到桑葚花色苷的最佳提取条件为:提取液为体积分数60%的酸化甲醇,超声温度41 ℃,超声时间39 min,料(g)液(mL)比1∶53。栽培蒙桑、野生蒙桑、黑果桑和白果桑桑葚的花色苷含量分别为11.815,11.166,9.179和0.189 mg/g,在花色苷质量浓度为0.1~2.0 mg/mL时,其氧自由基清除能力分别为0.100~0.666,0.100~0.500,0.115~0.615和0.029~0.441,还原能力分别为0.028~0.453,0.031~1.102,0.023~0.676和0.013~0.392,弱于相同质量浓度VC的氧自由基清除能力和还原能力。【结论】 4种桑葚中,栽培蒙桑的花色苷含量最高,氧清除能力最强,有进一步培育的价值。

Extraction of anthocyanins from four mulberry fruits by ultrasonic method and comparison of their antioxidative activities

Abstract:
【Objective】This study extracted anthocyanins from black fruit mulberry,white fruit mulberry,wild Morus mongolica and cultivated Morus mongolica by ultrasonic method and analyzed their contents and antioxidative activities to provide reference for breeding of mulberry species with high anthocyanin content and antioxidant ability in Jilin.【Method】The optimum extraction conditions were determined by response surface method.The anthocyanin was purified by AB 8 macroporous resins.Then,the free radical scavenging ability and reduction ability were determined and compared with VC.【Result】The optimum extraction conditions of anthocyanin determined by response surface method were:ethanol volume fraction 60%,ultrasonic temperature 41 ℃,ultrasonic time 39 min,and ratio of material (g) to liquid (mL) 1∶53.The anthocyanin contents of cultivated Morus mongolica,wild Morus mongolica,black fruit mulberry,and white fruit mulberry were 11.815,11.166,9.179,and 0.189 mg/g,respectively.When the concentration of anthocyanin concentration was 0.1-2.0 mg/mL,their superoxide free radical scavenging abilities were 0.100-0.666,0.100-0.500,0.115-0.615,and 0.029-0.441 and reduction abilities were 0.028-0.453,0.031-1.102,0.023-0.676,and 0.013-0.392,respectively.The abilities of superoxide free radical scavenging and reduction of four mulberry fruits were weaker than VC.【Conclusion】Cultivated Morus mongolica had the highest anthocyanin content and strongest free radical scavenging capacity.
