夏枯草水提物对肝损伤小鼠的保护作用

摘要:
【目的】研究夏枯草水提物对CCl4所致慢性肝损伤小鼠的保护作用。【方法】制备1 g/mL(以生药计)的夏枯草水提物,用不同剂量的夏枯草水提物(0.5,1,2 g/kg)每日灌胃小鼠,共5周,设立空白组和模型组,每日灌胃相同体积的生理盐水。除空白组注射植物油外,其余4组小鼠按5 mL/kg剂量腹腔注射体积分数1% CCl4植物油溶液,每周2次共3周,建立慢性肝损伤模型。于末次灌胃16 h后采血,测定血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性和丙二醛(MDA)浓度,并取肝脏样品制备肝组织匀浆,测定其中乳酸脱氢酶(LDH)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活性和总抗氧化能力(T-AOC),同时制备肝脏病理切片,观察肝组织病理组织学变化。【结果】与模型组相比,夏枯草水提物可明显降低血清ALT和AST活性、MDA浓度,提高肝组织匀浆中CAT、SOD活性以及T-AOC,降低LDH活性。显微观察结果显示,模型组小鼠肝小叶结构严重破坏,未见肝索和肝窦,肝细胞普遍变性;而夏枯草水提物各剂量组小鼠肝组织病灶逐渐缩小,病灶区部分肝细胞变性现象逐渐好转,肝索、肝窦结构逐渐出现。【结论】夏枯草水提物对CCl4所致慢性肝损伤具有明显的保护作用,其作用机制可能与抗氧化作用有关。

Abstract:
【Objective】This study investigated the protective effects of aqueous extract of prunella vulgaris L.on chronic hepatic injured mice induced by CCl4.【Method】Water extract of prunella vulgaris L.with concentration of 1 g/mL (in crude drugs) was prepared and administrated by gastric lavage to mice at different doses (0.5,1,and 2 g/kg) for 5 weeks,set up the blank group and model group,its daily lavage the same volume of physiological saline.Four groups of mice were administrated with volume fraction 1% CCl4 vegetable oil solution (5 mL/kg body weight,intraperitoneal injection twice each week for 3 weeks) to establish the chronic hepatic injury model while the control mice were given vegetable oil.16 h after the last intragastric administration,blood was collected and the activities of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) as well as serum malondialdehyde (MDA) concentration were determined.Liver tissues were also homogenized to measure the activities of hepatic lactate dehydrogenase (LDH),catalase (CAT),and superoxide dismutase (SOD),as well as total antioxidant capacity (T-AOC) level.Moreover,liver sections were prepared to observe histopathological changes in tissue.【Result】Compared with the model group,water extract of prunella vulgaris L.significantly decreased the activities of serum ALT and AST and serum MDA concentration,increased the activities of hepatic CAT and SOD and hepatic T-AOC,and reduced the activity of hepatic LDH.Microscopic observation showed that the hepatic lobule structure of control group mice was severely damaged,no hepatic cords and hepatic sinusoids were found,and hepatic cells were generally denatured.Different dose groups of water extract of prunella vulgaris L.gradually reduced the focus of mice liver tissue,the degeneration phenomena of liver cells in focus was gradually improved,and the structure of liver cords and liver sinusoids gradually appeared again.【Conclusion】Water extract of prunella vulgaris L.exerted protective effects on chronic hepatic injured mice induced by CCl4,and the mechanisms may be related to its antioxidant effect.
