甘蓝小孢子胚状体分化不定芽再生植株的研究

摘要:
【目的】探讨甘蓝小孢子胚状体诱导不定芽的适宜生根转接高度及添加烯效唑对再生植株生长的影响,寻找胚状体芽生根和再生植株生长发育的最佳培养条件,为甘蓝小孢子培养技术体系的不断完善奠定基础。【方法】以“绿球66”甘蓝游离小孢子为材料,诱导分化出不定芽,根据芽高度不同分为≥1.0~<2.0 cm (小芽)和≥2.0~<3.5 cm (大芽) 2组,分别转接到MS+NAA 0.2 mg/L+烯效唑(0(对照),0.01,0.05,0.1 mg/L)+Suc 30 g/L+Agar 7 g/L培养基上诱导生根,培养再生植株,观测再生植株的生长情况,筛选不定芽适宜转接高度和烯效唑最佳质量浓度。【结果】甘蓝小孢子胚状体诱导生成不定芽高度≥1.0~<3.5 cm时转接生根,不仅能促进胚状体丛生芽中小芽继续生长,而且有利于增加当代小孢子再生植株数量。对照甘蓝小孢子再生植株根系根瘦弱细长、数量少、韧性较差;添加0.01和0.05 mg/L烯效唑后,再生植株根短粗、数量多、韧性强、健壮,且以0.05 mg/L烯效唑处理的再生植株生长最好。生根培养基中添加0.01和0.05 mg/L烯效唑后,小芽再生植株移栽到营养钵和定植到田间的成活率分别为90.7%,98.8%和89.7%,97.6%,大芽再生植株则分别为91.0%,100.0%和94.4%,97.7%。【结论】甘蓝小孢子在MS+NAA 0.2 mg/L+烯效唑0.05 mg/L+ Suc 30 g/L+Agar 7 g/L培养基中生根再生的植株,其移栽到营养钵和田间的成活率均超过了97.6%。

Adventitious bud regeneration plant differentiation from cabbage microspore embryoid
Abstract:
【Objective】This article discussed the effects of root switching height and uniconazole on adventitious bud induced by cabbage microspore embryo to obtain the best cultivation condition.【Method】Cabbage cultivar “Lvqiu66” was selected as the material.According to bud height,the cultivars were divided into ≥1.0-<2.0 cm group (small bud) and ≥2.0-<3.5 cm (big bud) group before being transferred to medium of MS+NAA 0.2 mg/L+uniconazole (0 (CK),0.01,0.05,0.1 mg/L)+Suc 30 g/L+Agar 7 g/L to induce root and culture regeneration plant.The growth conditions of adventitious bud were observed and the best concentration of uniconazde was analyzed.【Result】The height of ≥1.0~<3.5 cm could promote the growth of cabbage microspore embryoids induced adventitious bud and significantly improve the number of regeneration plants.Regeneration plant root system of control was thin slender with less number and poor toughness while adding uniconazole improved the quantity,toughness and robustness.0.05 mg/L uniconazole was the best treatment.The transplanting survival rates and engraftment rates of regeneration plants of small bud group were 90.7%,98.8% and 89.7%,97.6%,respectively and that of big bud group were 91.0%,100.0% and 94.4%,97.7%,respectively.【Conclusion】Regeneration plants of cabbage microspore in the medium of MS+NAA 0.2 mg/L+uniconazole 0.05 mg/L+Suc 30 g/L+Agar 7 g/L were the best with transplanting survival and engraftment rates of higher than 97.6%.
