醋酸铅试纸法在低产H2S酿酒酵母筛选中的应用

摘要:
[目的]探讨醋酸铅试纸法筛选低产硫化氢(H2S)酿酒酵母的可行性.[方法]在模拟汁培养基发酵条件下,以采自新疆鄯善地区酿酒葡萄自然发酵液中的10株野生型酿酒酵母(M1,M2,…,M10)为对象,采用醋酸铅试纸法对其中的低产H2S酵母进行了初筛和复筛.[结果]经醋酸铅试纸初筛后,菌株M2、M3、M6属于低产H2S的菌株,而M1、M8、M10属于高产H2S的菌株.复筛结果显示,菌株M2、M3、M6的硫化氢生成量较低,均为12 μg/L;菌株M10的H2S生成量最高,为1 920 μg/L.模拟发酵试验表明,有8株野生型酿酒酵母(M7和M9除外)发酵正常,具备良好的发酵能力,酒精度都在10%(体积分数)以上.[结论]醋酸铅试纸法可用于低产H2S酿酒酵母的筛选,该法具有结果稳定,方法简便,成本低廉,操作简单等优点,有一定的推广价值.菌株M2、M3和M6皆为低产H2S酿酒酵母,且其发酵能力适合工业发酵要求.

Application of lead acetate test paper in the screen of low-H2S-producing yeast strains

Abstract:
【Objective】 The feasibility of the lead acetate test paper in screening of low-yield H2S yeast strains was studied.【Method】 With 10 wild Saccharomyces cerevisiae yeast strains,separated from Shanshan,Xinjiang province,as subject,low-yield H2S yeast strains were screened twice in the fermented simulated juice with the method of lead acetate test paper.【Result】 The results showed that M2,M3,M6 were low-yield H2S strains,M1,M8,M10 high-yield H2S strains.Among them,M2,M3,M6 produced H2S 12 μg/L;M10 could produce the highest content of H2S by 1920 μg/L.With higher ethanol-production ability (all above 10%) 8 wild yeast strains (except M7 and M9) showed superior characters in the fermentation experiment.【Conclusion】 The method of lead acetate test paper is convenient,low-cost,stable,and has high value for expansion and application in the screen of low-yield H2S yeast strains.With the method,the three yeast strains suitable for industrial fermentation of M2,M3,M6 were screened as low-yield H2S strains.
