苦马豆素降解酶的酶促降解特性

摘要:
[目的]研究苦马豆素(Swainsonine, SW)降解酶的酶促降解特性.[方法]应用渗透休克原理,提取SW降解菌株YLZZ-1粗酶液,进行酶促降解研究,并考察SW降解酶的酶活力、动力学参数及稳定性.[结果]YLZZ-1菌株的胞内粗酶液对SW的降解率高达91.73%.SW降解酶对SW酶促降解的最适pH为8.0,最适温度为30℃,在pH6.0~8.5、温度25~45℃具有较好的降解活性,其米氏常数Km为0.073 98 mmol/L,最大反应速度Vmax为0.111 5 μmol/(mg·min).SW降解酶有较好的pH稳定性,但对热不稳定.[结论]菌株YLZZ-1的SW降解酶属于胞内诱导酶,具有良好的降解能力.

Abstract:
【Objective】 Degradation characteristics of swainsonine by the crude enzyme of strain YLZZ 1was investigated.【Method】 Crude enzyme of YLZZ-1 was extracted through osmotic shock principle.The enzyme activity,dynamic parameter and stability of the main catabolic enzymes were researched.【Result】 The degradation rate for SW by intracellular enzyme was 91.73%.The pH optimum for crude enzyme was 8.0 for enzymatic degradation of SW,and it had comparatively high activity in the range of pH 6.0-8.5.The optimum temperature for enzymatic activity was 30 ℃,and it still had comparatively high activity in the range of temperature 25-45 ℃.The crude enzyme showingKm value was SW 0.073 98 mmol/L,and the maximal enzymatic degradation rate was 0.1115 μmoL/(mgomin).The enzyme had the stable pH,but was sensitive to high temperature.【Conclusion】 The intracellular and induced enzyme was the main SW catabolic enzymes of YLZZ-1,and it had excellent degradation ability.
