[目的]构建小鼠早期胚胎与皮肤成纤维细胞共生的微环境体系.[方法]分离培养EGFP纯合阳性成年ICR小鼠皮肤成纤维细胞.取激素超排合笼后2.5 d小鼠8-细胞胚胎,通过显微操作系统将4~6个EGFP阳性小鼠皮肤成纤维细胞注射到小鼠早期8-细胞胚胎中,转移发育到囊胚阶段的共生胚胎至小鼠胎儿成纤维细胞饲养层上,在小鼠胚胎干细胞分离培养条件下进行培养.[结果] EGFP阳性小鼠成纤维细胞注射到小鼠胚胎后,荧光显微镜下显示,EGFP阳性小鼠成纤维细胞分布到小鼠囊胚组成细胞中,参与小鼠囊胚组成.在发育到囊胚的共生胚胎中,EGFP阳性小鼠成纤维细胞分布到小鼠囊胚细胞中的比例为93.73%(269/287).在小鼠胚胎干细胞的分离培养条件下,2.60%(7/269)共生胚胎中EGFP阳性小鼠成纤维细胞参与小鼠胚胎内细胞团的组成.[结论]小鼠皮肤成纤维细胞与小鼠早期胚胎能够形成共生的微环境体系. |

Abstract: |
【Objective】 Embryos with early mouse embryo and adult cells were made to research the growth and development of adult cells in embryonic microenvironment. 【Method】 EGFP positive fibroblasts from mouse skin were isolated and cultured, and then injected into early mouse 8-cell embryos by micromanipulation.Symbiotic blastocysts from mouse fibroblasts and 8-cell embryos were transferred onto mouse fetal fibroblast feeder cells with mouse embryonic stem cell isolation and culture media. 【Result】 93.73%(269/287) symbiotic blastocysts were successfully constructed with EGFP positive mouse fibroblasts and 8-cell embryos,EGFP positive mouse fibroblasts became a member of symbiotic blastocysts and were distributed into different parts of symbiotic blastocysts with fluorescence microscope.In addition,the images of EGFP positive mouse fibroblasts were changed and EGFP positive fibroblasts took part in inner mass cell in 2.60%(7/269) symbiotic blastocysts. 【Conclusion】 Cells in adult mice with early mouse embryos can form a symbiotic microenvironment system. |
