为了考察大孔吸附树脂对苦参水提液中氧化苦参碱的分离效果,选择5种大孔吸附树脂,并采用HPLC定量分析法,研究了不同大孔吸附树脂对氧化苦参碱的吸附性能。结果表明,大孔吸附树脂LSA-21和LSA-30的分离效果明显优于LSA-40,HP-10和AB-8;LSA-21在pH10.0和0 ℃时吸附效果最好,其动态饱和吸附率为14.52 mg/mL,静态饱和吸附率为6.88 mg/mL,最适洗脱剂为体积分数30%乙醇,解吸率可达90%以上,由此得到的氧化苦参碱含量约为37%。静态饱和吸附率随温度的升高而降低,其中在较低温度范围(0~10 ℃)内影响显著;静态吸附速率随时间的延长而变小,其中在吸附的前2 h内,吸附速率很大且基本恒定,2 h之后吸附速率变得非常小。 |

Separation of oxymatrine from Sophora flavescens by macroporous absorption resin |

Abstract: |
The separation effects of oxymatrine from Sophora flavescens with five types of macroporous absorption resins were studied.The results showed that macroporous absorption resin LSA-21 and LSA 30 were much better than LSA-40,HP-10 and AB-8 in absorption ratio;The optimum absorption conditions of LSA-21 were pH 10.0 and 0 ℃,and its maximum dynamic absorption ratio and maximum static absorption ratio were 14.52 mg/mL and 6.88 mg/mL respectively,ideal eluent was 30% ethanol,which led to ≥90% de-absorption ratio and about 37% oxymatrine in dry eluate;The static absorption ratio would drop as the temperature rose and was more obvious in lower temperature range(0-10 ℃),and meanwhile the static absorption speed slowed with the prolonging of time,the absorption ratio being fast and constant in the first 2 hours,then much slower. |
