中国牦牛β-珠蛋白基因的克隆和序列分析

摘要:
根据黄牛β-珠蛋白基因的序列设计引物,扩增了中国牦牛的β-珠蛋白基因.并对其进行了克隆测序和氨基酸序列比较分析。结果显示,其与黄牛β-珠蛋白基因的同源性很高.达到97%以上;检测到中国牦牛β-珠蛋白基因的2个等位基因β73Asn和β135Asn,其中67号牦牛个体中检测到2个等位基因β73Asn和β135Asn,64号牦牛个休中检测到等位基因β135Asn,1078号牦牛个体中检测到等位基困β73Asn;等位基因β135Asn是中国牦牛特有的一个等位基因,推测该等位基因编码的第135位氨基酸天冬酰胺,可能会降低牦牛血红蛋白的氧亲和力。

Abstract:
Yak β-globin is amplified by primers designed from cattle β-globin gene.The cloning and sequencing results indicate that they have very high homology with cattle β-globin gene with a value of above 97%.Two alleles,β73Asn and β135Asn,were detected at β-globin locus in Chinese yak.The sample 67 has these two alleles,while individual 64 has allele β135Asn and individual 1078 has allele β73Asn.The allele β135Asn is the characteristic of Chinese yak after residue blasting.It is suggested that the β135 Asn of this allele could reduce the oxygen affinity of yak hemoglobin.
