提高八楞海棠遗传转化植株再生率技术的研究

摘要:
为进一步提高八楞海棠遗传转化效率,以八楞海棠试管苗不同组织部位(叶片、叶柄、节间茎段、破坏生长点的茎尖)为外植体,在含有0,1,3,5,10 mg/L除草剂草丁膦(glufosinate)的分化培养基(MS+4 mg/L 6-BA +0.2 mg/L NAA)上进行筛选压力的确定,其中3 mg/L除草剂为适宜的筛选压力;在此筛选压力下,以八楞海棠不同外植体进行遗传转化,其中破坏生长点的茎尖处理组获得的再生芽效率最高,通过对部分再生苗进行PCR及Southern杂交检测,均鉴定为转化苗。

Study on enhancing efficiency of Malus robusta transformation
Abstract:
In order to enhance efficiency of Malus robusta gene transformation,different parts of Malus robusta plantnets were used as explants to determine screening stress on the development media (MS+4 mg/L 6-BA+0.2 mg/L NAA) containing 0,1,3,5,10 mg/L glufosinate,of which 3 mg/L was the acceptable screening stress.The gene transformation was made by using M.robusta explants under screening stress.Regeneration rates of those shoots with destroyed growth points were the highest.Some generation plants were confirmed to be transformation plants by PCR and Southern crossing analysis.
