以日本木立芦荟(A.arborescens Mill.)茎尖和幼嫩茎段为外植体进行离体快繁。结果表明,茎尖在MS +6-BA 3.0 mg/L +NAA 0.2 mg/L培养基上形成的不定芽数最多,达21个;而茎段在MS +6-BA 4.0 mg/L +NAA 0.1 mg/L培养基上诱导的不定芽数最多,为8.1个。MS +6-BA 3.0 mg/L +B9 100 mg/L培养基对不定芽的增殖效果最好,平均每个芽可增殖不定芽11.5个。1.0mg/L NAA和2.0 mg/L IBA能显著促进不定芽生根,生长延缓剂CCC也具有促进生根作用。将组培苗移栽到体积比组成为2份有机厩肥 +1份园田土或2份蛭石 +1份园田土的基质中,成活率可达90%以上。 |

Abstract: |
Tissue culture of A.arborescens was studied with meristem and young stem segments as explants.The optimum media for induction of adventitious buds from meristem and stem segment were MS+6-BA 3.0 mg/L+NAA 0.2 mg/L and MS+6-BA 4.0 mg/L+NAA 0.1 mg/L respectively with the bud number reaching to 21 and 8.1.Shoots were propagated as much as 11.5 when they were placed at MS+6-BA 3.0 mg/L + B9(duminozide) 100 mg/L.1.0 mg/L NAA and 2.0 mg/L IBA were the best treatments for the induction of roots.Besides NAA and IBA,CCC a kind of inhibitor for growth also was helpful to promote shoot to root differentiation.More than 90% of the shoots with roots survived after planted into mixture of 2 stable manure+1 soil or 2 vermiculite+1 soil. |
