牛卵泡卵母细胞冷冻保存的研究

摘要:
研究了用3种不同方法(A法:程序冷冻;B法:超快速冷冻;C法:玻璃化冷冻)冷冻保存的牛GV期卵母细胞的发育潜力。结果表明,解 冻后,C组卵母细胞形态正常率显著高于A、B组;3种方法冷冻的卵母细胞成熟培养后,平均卵丘扩展率为65%,其中C组最高(69%);3组冷冻卵母细胞的体外成熟率、体外受精率和卵裂率分别为14.3%,17.1%,21.6%;11.1%,12.6%,15.1%和5.6%,9.1%,11.1%。结果还表明:玻璃化冷冻的卵母细胞损伤较轻。

Research on cryopreservation of bovine follicular oocytes
Abstract:
The developmental competences of bovine GV-Stage oocytes cryopreserved by three methods (Method A:Conventional freezing procedures;Method B:ultrarapid freezing;Method C:Vitrification) were studied.After thawing,the morphological normal rate of C gronp was significantly higher than those of group A and B.The average cumulus expansion rate of all the cultured oocytes was 65% and that of group C was the highest(69%).The in vitro maturation rates were 14.3%,17.1% and 21.6%,and the fertilization rates were 11.1%,12.5% and 15.0%,and the oocyte cleavage rates were 5.6%,9.1% and 11.1% in group A,B,C oocytes cryopreserved respectively.The results showed that the injuries of vitrified oocytes were less severe.
